山奈酚对小鼠黑素瘤细胞DNA损伤的影响

摘　　要：目的探讨山奈酚对小鼠黑素瘤细胞B16 DNA损伤的影响。方法体外实验将不同浓度的山奈酚作用于B16细胞,通过DAPI染色荧光倒置显微镜观察细胞凋亡形态,彗星电泳检测分析山奈酚对B16细胞DNA的损伤情况,流式细胞术检测细胞凋亡率。结果荧光倒置显微镜下显示当细胞经12.5μmol/L、25μmol/L、50μmol/L的山奈酚处理72h后,与阴性对照组比较,细胞出现明显的凋亡形态,且随着药物浓度的增高,凋亡现象越明显。彗星电泳检测经12.5μmol/L、25μmol/L、50μmol/L山奈酚作用72h的B16细胞,与阴性对照组比较,细胞核平均光密度值减小,彗星尾距延长,且随着药物浓度的升高二者改变越明显,呈现量效关系,各药物组数值与阴性对照组比较差异有统计学意义(P<0.01)。流式细胞术检测经12.5、25、50μmol/L的山奈酚处理72h后的B16细胞,与阴性对照组比较,凋亡率增加。结论体外实验山奈酚对小鼠黑素瘤细胞B16 DNA具有损伤作用。Objective To explore the effects of kaempferol to DNA on B16 murine melanoma cells.Methods We detected the activity of kaempferol to DNA through DAPI stain assay,comet assay and flow cytometry re- spectively in vitro experiments.Results Treated by 12.5,25,50μmol/L kaempferol respectively 72hours later in vitro, the cells showed the characters of apoptosis and significant dose- effect relationship.The comet assay of B16 treated with 12.5,25,50μmol/L kaempferol respectively 72hours illustrated that the average light den- sity declined and the back range increased versus negative control, with obvious dose - effect relationship, Prob - value was less than 0.01 versus negative control.The flow cytometry assay illustrated that the apoptosis rate of B16 treated with 12.5,25,50μmol/L kaempferol significantly increased versus control group.Conclusion Kaempferol can induce DNA damage on B16 routine melanoma cells.
