右美托咪定对烧伤大鼠核转录因子kappaB和肿瘤坏死因子-α的影响

摘　　要：目的观察右美托咪定预处理对严重烧伤大鼠表达核转录因子kappa B(nuclear factor-kappa B,NF-κB)和肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)的影响。方法成年健康SD大鼠128只,随机分为4组:假手术组(S组),烧伤对照组(C组),右美托咪定组(D组),右美托咪定+育亨宾组(D+Y组),每组32只。分别收集烧伤后2h,6h,12h和24h的血清,ELISA检测各组大鼠血清内NF-κB和TNF-α的表达变化情况。结果烫伤后的C组大鼠,随着损伤时间的延长,体内血清含有的NF-κB和TNF-α逐步增高,在伤后高于相对应的S组。D组NF-κB在伤后6h和C组比较,有下调(P<0.05),D+Y组在伤后12h和24h NF-κB的含量高于D组(P<0.05),但和C组比较差异无统计学意义(P>0.05);D组TNF-α在伤后12h较C组有下调(P<0.05),D+Y组在伤后12h和24h TNF-α的表达高于D组(P<0.05),但和C组比较差异无统计学意义(P>0.05)。结论通过右美托咪定预处理后,严重烧伤大鼠体内NF-κB和TNF-a的表达降低,有助于减轻严重烧伤的炎症反应。Objective To observe the effects of dexmedetomidine preconditioning on expression of NF- kap- pa B and TNF- alpha in severely burned rats.Methods Totally 128 SD rats were randomly divided into 4 groups ： sham operation group （group S） , control group （group C） and dexmedetomidine group （group D） , dexmedetomidine ＋ yohimbine group （group D＋ Y） ,32 rats in each group.The serum was collected at 2h, 12h,6h and 24h after burn. The expression NF-κB and TNF-？ was detected by ELISA. Results In the group C,the levels of NF- κB and TNF- α were increased gradually,and the group C was significantly higher than the group S after burn. Compared with group C, NF- κB expression in group D was significantly de- creased at 6h after burn（P〈0.05） ,the content of NF- κB in group D＋ Y was higher than group D at 12h and 24h after burn （P〈0.05） ,but there was no significant difference between group D and group C （P〉0.05）. Compared with C group,TNF-α expression in group D was significantly decreased at 12h after burn（ P 〈0.05） ,the expression of TNF- α in group D ＋ Y was higher than group D at 12h and 24h after burn （P〈 0.05） ,but there was no significant difference between group D and group C （P〉0.05）.Conclusion After pre- treatment with dexmedetomidine, the expression of NF - κB and TNF - a in severely burned rats is decreased, which is helpful to alleviate the inflammatory reaction.
