摘  要:目的评价阿托伐他汀预处理对异氟醚麻醉小鼠认知功能的影响。 方法健康雄性C57BL/6小鼠48只,3月龄,体重27~41 g,采用随机数字表法分为3组(n=16):对照组(C组)、异氟醚麻醉组(I组)和阿托伐他汀预处理+异氟醚麻醉组(AI组)。AI组小鼠于每天同一时点给予阿托伐他汀10 mg/kg灌胃,连续给药7 d。I组及AI组于给药结束后1 d,持续吸入1.5%异氟醚6 h,吸入氧流量2 L/min。于麻醉结束后1 d行旷场实验及水迷宫实验。于水迷宫实验结束后1 d处死小鼠,取海马组织,采用Western blot法检测caspase-3、Bax和Bcl-2的表达水平,采用ELISA法检测IL-1β、TNF-α和可溶性Aβ1-42的含量。 结果各组小鼠旷场实验各指标比较差异无统计学意义(P〉0.05)。与C组比较,I组和AI组各时点逃避潜伏期延长,原平台所在象限停留时间缩短,穿越原平台次数减少,海马组织IL-1β、TNF-α和可溶性Aβ1-42含量升高,caspase-3和Bax表达上调,Bcl-2表达下调(P〈0.05);与I组比较,AI组各时点逃避潜伏期缩短,原平台所在象限停留时间延长,穿越原平台次数增加,海马组织IL-1β、TNF-α和可溶性Aβ1-42含量降低,caspase-3和Bax表达下调,Bcl-2表达上调(P〈0.05)。 结论阿托伐他汀预处理可改善异氟醚麻醉小鼠认知功能,其机制可能与减轻海马炎症反应,抑制Aβ1-42过度表达,抑制神经元凋亡有关。Objective To evaluate the effect of atorvastatin preconditioning on cognitive function in isoflurane-anaesthetized mice. Methods Forty-eight healthy male C57BL/6 mice, aged 3 months, weig- hing 27-41 g, were divided into 3 groups (n= 16 each) using a random number table: control group (group C) , isoflurane anesthesia group (group D and atorvastatin preconditioning plus isoflurane anesthe- sia group (group AI). Atorvastatin 10 mg/kg was given through a gastric tube into the stomach at the same time every day for 7 consecutive days in group AI. In I and AI groups, 1.5% isoflurane was inhaled for 6 h with fresh gas flow of 2 L/rain at 1 day after the end of administration. Open field test and Morris water maze test were performed at 1 day after the end of anesthesia. The mice were sacrificed at 1 day after the end of Morris water maze test, and hippocampi were isolated for determination of caspase-3, Bax and Bel-2 ex- pression ( by Western blot) and contents of interleukin- lbeta ( IL- 1β) , tumor necrosis factor-alpha ( TNF- a) and soluble A: in hippocampal tissues (by enzyme-linked iminunosorbent assay). Results There was no significant difference in the parameters of open field test among the three groups (P〉0.05). Com- pared with group C, the escape latency was significantly prolonged at each time point, the time of staying at the original platform quadrant was shortened, the frequency of crossing the original platform was decreased, the contents of IL-1β, TNF-a and soluble A[5.42 were increased, the expression of caspase-3 and Bax wasup-regulated, and Bcl-2 expression was down-regulated in I and AI groups (P〈 0.05). Compared with group I, the escape latency was significantly shortened at each time point, the time of staying at the original platform quadrant was prolonged, the frequency of crossing the original platform was increased, the con- tents of IL-113, TNF-a and soluble A1_42 were decreased, the expression of caspase-3 and Bax was down- regulated, and Bcl-
