摘  要:目的建立HPLC分析方法测定危重症患者行连续性肾脏替代治疗超滤液中奥硝唑的含量,监测连续性肾脏替代治疗对抗生素类药物代谢的影响,为临床个体化给药提供依据,合理使用抗生素。方法色谱柱为Agilent ZORBAX SBC_(18)(4.6mm×150mm,5μm);流动相为甲醇-水(30:70,V/V),流速为1.0m L/min;检测波长为318nm;柱温为25?C;进样量为10μL,运行时间9min。结果奥硝唑在2.00~40.00μg/m L范围内与其峰面积线性关系良好(r=1.000),平均回收率为98.84%(n=9),RSD为0.87%。结论该方法简单、快速,结果准确且重复性好,可用于连续性肾脏替代治疗超滤液中奥硝唑含量的测定,为奥硝唑的临床研究提供可靠的分析方法。Objective To establish an HPLC method for the determination of omidazole in the ultra-filtrate liquid of the continuous renal replacement therapy (CRRT) for critically ill patients,to monitor the effects of CRRT ultra-filtrate liquid on the metabolism of antibiotics,and to provide a basis for the individualized medication and reasonable use of antibiotics.Methods An agilent ZORBAX SB-C18 (4.6mm×150mm,5μm) was chosen for the analysis.The mobile phase consisted of methanol and water (30:70,V/V).The flow rate was 1.0mL/min with the detection wavelength of 318nm,the column temperature was 25 ℃,the injection volume was 10μL,and the running time was 9min.Results A good linear correlation of omidazole was observed within the range of 2.00~40.00μg/ mL (r=l.000).The average recovery rate of ornidazole was 98.84% (n=9),with RSD of 0.87%.Conclusion This method was rapid,convenient,accurate,robust and suitable for the determination of ornidazole in CRRT ultra-filtrate liquid.A reliable analytical method was provided for the clinical study of omidazole.
