摘 要:目的观察香茅精油对D-半乳糖联合三氯化铝(Al Cl3)致小鼠老年痴呆样损伤的影响并研究可能的作用机制。方法选50只昆明小鼠,皮下注射5%D-半乳糖120 mg·kg^(-1)+灌胃Al Cl3200 mg·kg^(-1)共10周,建立小鼠老年痴呆模型。另选10只作为正常组。按照体重将造模成功的老年痴呆小鼠随机分为5组:模型组、对照组(脑复康0.5 g·kg^(-1)灌胃)、低、中、高3个剂量实验组(香茅精油0.4,0.6,0.8 m L·kg^(-1)灌胃),各组灌胃体积均为10μL·g^(-1),每日1次,从造模第11周开始给药,连续给药4周。正常组与模型组灌胃0.9%Na Cl。用Morris水迷宫测定小鼠的学习记忆能力,以分光光度法检测血清的丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)和脑组织的乙酰胆碱(ACh)含量。结果训练至第5天,正常组、模型组、对照组与低、中、高3个剂量实验组的逃避潜伏期(s)分别为21.42±3.18,43.33±2.14,26.84±0.71,23.16±3.70,18.48±1.11,21.40±0.88。与正常组相比,模型组差异有统计学意义(P<0.01)。与模型组比较,对照组与低、中、高3个剂量实验组差异均有统计学意义(P<0.05或0.01)。正常组、模型组、对照组与低、中、高3个剂量实验组的血清GSH-PX含量分别(299.05±90.62),(246.19±37.90),(532.49±121.52),(587.21±177.37),(497.14±82.95),(493.33±73.64)U·m L^(-1)。这6组的血清MDA含量分别(9.38±3.01),(14.89±4.41),(6.98±2.43),(6.06±3.34),(5.26±1.90),(5.01±1.59)mmol·m L^(-1)。与正常组比较,模型组差异均有统计学意义(均P<0.05);与模型组比较,这6组差异均有统计学意义(P<0.05或P<0.01)。正常组、模型组、对照组与低、中、高3个剂量实验组的脑组织中ACh含量分别为(268.27±25.40),(224.95±21.52),(392.56±50.58),(240.17±17.15),(255.50±15.91),(259.15±20.74)μg·m L-1。与正常组比较,模型组差异有统计学意义(P<0.01);与模型组比较,对照组与中高2个剂量实验组差异均有统计学意义(P<0.05,P<0.01)。结论香茅精Objective To investigate the effect of Citronella oil on dementia of Alzheimer' s type induced by D-galactose and aluminum trichloride( AlCl3) and explore its mechanism. Methods The 60 healthy KM mice with sex in half were enrolled in this study,10 of them were taken randomly as the normal group,the remainder 50 mice were in the model group. The rats received inntraperitoneal injection of D-galactoseand oral aluminum trichloride for consecutive 10 weeks to construct the disease models. The model rats were randomly divided into five groups: model group,control group( 0. 5 g · kg-1 Piracetan) and three doses( 0. 4,0. 6,0. 8 m L·kg-1 Citronella oil) experimental groups. The ability of learning and memory was measured by the Morris water maze test. The levels of glutathione peroxidase( GSH-PX) and malondialdehyde( MDA) in serum homogenate and acetylcholine( ACh) in brain tissue homogenate were determined by spectrophotometer. Results At the fifth day in the Morris water maze training period,the escape latency( EL; s) in the normal group,model group,control group and three doses experimental groups were 21. 42 ± 3. 18,43. 33 ± 2. 14,26. 84 ± 0. 71,23. 16 ± 3. 70,18. 48 ± 1. 11,21. 40 ± 0. 88. Compared with the normal group,the EL in model group was increased significantly( P〈0. 05).Compared with the model group,EL in control group and three doses experimental groups reduced significantly( P〈0. 05).
