摘 要:目的探讨"双固一通"电针对亚急性衰老模型大鼠脑部海马基因表达及JAK/STAT信号通路的影响。方法选用SD大鼠随机分为正常对照组(正常组)、亚急性衰老模型组(模型组)、"双固一通"电针组(电针组),每组10只。除正常组外,其余两组采用D-半乳糖腹腔注射制作亚急性衰老模型。电针组予以"双固一通"电针治疗,每日1次,每周治疗6次,共治疗4周。采用Morris水迷宫检测各组大鼠的学习记忆能力;酶标仪检测大鼠血清中超氧化物歧化酶(SOD)的含量。基因芯片技术测定大鼠脑部海马区的基因表达,并进一步通过RT-PCR技术验证差异基因pth2r、JAK1、JAK2、STAT4的表达。结果与正常组比较,模型组大鼠逃避潜伏期、首次跨越原平台时间、跨越次数均显示显著差异(P<0.01),血清SOD活性也显著降低(P<0.01);与模型组比较,电针组大鼠逃避潜伏期、首次跨越原平台时间、跨越次数均显示显著差异(P<0.01),血清SOD活性明显升高(P<0.01)。采用illumina基因谱芯片对三组大鼠脑部海马组织进行检测分析。以差异基因pth2r为参照进行RT-PCR验证表明,JAK1、JAK2、STAT4表达与基因芯片结果一致,与正常组比较,模型组海马JAK1、JAK2基因相对高表达(P<0.01)、STAT4基因低表达(P<0.01);而经电针处理的海马JAK1、JAK2基因的表达量下降(P<0.01)、STAT4基因表达量明显升高(P<0.01)。结论"双固一通"电针改善亚急性衰老模型大鼠的学习记忆能力,与提高血清SOD活性和对海马基因表达及JAK/STAT信号通路的影响相关。Objective To explore the influence of "Shuanggu Yitong" electro-acupuncture effects on the gene expressin and JAK/STAT signaling pathway to the hippocampus area in sub- acutely aging rats. Methods 30 Sprague- Dawley rats were randomly divided into three groups: normal group(NG), sub-acute aging model group(MG)and"Shuanggu Yitong" electro- acupuncture group(AEG'), 10 for each group. Rats in NG and MG were obtained by D- galactose intraperitoneal injection con- tinuously for 40 days. The acupoints "housanli ( ST 36 )", "Guanyuan ( CV 4 )" and "Baihui ( DU 20 )" were used for electro- acupuncture treatment. EA treatment was performed every day except Sunday and lasted for 4 weeks. Morris water maze was carried out to test the learning and memory ability of rats in different groups. The serum SOD contents of rats in different groups were tested by ELISA assay, cDNA microarray was used to explore the gene expression pattern of genes in the hippocampus tissue. RT- PCR was used to test the expression level of differentially expressed genes. Results Compared with NG, rats in MG showed decreased learning and memory ability and the serum SOD content(P〈0.01 ). Compared with MG rats, the learning and memory ability and the serum SOD content were increased in AEG rats. 4052 DE genes were identified in the cDNA microarray test. 3079 DE genes were identified between NG and MG, and 983 DE genes were identified between AEG and MG. The RT- PCR result showed the expression of genes pth2r, JAK1, JAK2, STAT4 were consistent with the cDNA microarray results. Conclusion "Shuanggu Yitong" electro-acupuncture is effective in improving the learning and memory ability for sub - acutely aging rats, it may be asSoCiated with the improving serum SOD content and the regulating of gene expression and JAK/ STAT signaling pathway to the hippocampus in subacute aging rats.
