摘  要:目的:研究慢性低O_2高CO_2对小鼠海马细胞凋亡、caspase-3以及p38 MAPK活性的影响。方法:雄性C57BL/6小鼠24只,随机分成2组:4HH组(低O_2高CO_2组12只)和NC组(正常对照组12只)。4HH组饲养于氧舱中,舱内O_2浓度为9%~11%,CO_2浓度为5%~6%,每天8 h,每周6 d,共4周,其余时间与NC组一样,生活于正常环境。原位末端标记(TUNEL)法检测海马细胞凋亡指数(AI),caspase-3活性检测试剂盒测定海马caspase-3活性,Western blot检测海马p-p38 MAPK蛋白的表达。结果:4HH组小鼠海马细胞凋亡指数(AI)、caspase-3活性及p-p38 MAPK蛋白含量显著高于NC组(P<0.01)。结论:慢性低O_2高CO_2促进小鼠海马细胞凋亡,促进caspase-3以及p38 MAPK活化。Objective: To investigate the effect of the chronic hypoxic-hypercapnia in apoptosis and activity of p38 MAPK and caspase-3 of hippocampus in mouse. Methods: Twenty-four male mice were randomly divided into two groups: the hypoxic-hypercapnia 4 weeks (4HH) group and normal control (NC) group. The 4HH groups in the special cantainer were exposed to an atmosphere containing 9% - 11% 02 and 5% - 6% CO2 8 hours a day, 6 days a week for 4 weeks. The two groups lived in the same room in the rest time. Apoptosis index (AI) in hip- pocampus was counted by TUNEL staining. Caspase-3 activity was measured by a caspase-3 activity assay kit. p-p38 MAPK protein in hippocampus was measured by Western blot. Results: Compared with NC group the AI, caspase-3 activity, and the p-p38 MAPK protein expression in hippocampus were significantly increased in 4HH group (P〈0.01). Conclusion: The chronic hypoxic-hypercapnia promotes apoptosis and the activation of cas- pase-3 and p38 MAPK of hippocampus in mouse.
