摘  要:目的探讨中度海拔地区高原低氧环境影响男性精液质量是否与Chk1表达变化有关。方法收集正常精子组、少精子组、弱精子组和少弱精子组的标本各20份,采用qRT-PCR和Western blot方法,从mRNA和蛋白水平检测各组精子中Chk1的表达,并对相关结果行统计学分析。结果 1)在各组精子中均可检测到Chk1蛋白的表达;2)少精子组、弱精子组和少弱精子组精子中Chk1mRNA及蛋白表达量显著低于正常精子组(P<0.05),且在少弱精子组表达最低。结论Chk1在中度海拔地区汉族人群男性不育患者精子中表达下调,可能与高原低氧环境影响精液质量有关。Objective To investigate whether the semen checkpoint kinase 1 ( Chkl ) expression affected by the hypoxia environment in the middle altitude area.Methods 80 semen samples were collectedand divided them into 4 groups : normal ( 20 ), oligozoospermia ( 20), asthenospermia ( 20 ) and oligoasthenozoospermia ( 20 ). The expression of Chkl mRNA and protein in sperm were detected by qRT-PCR and Western blot.Results 1) Sperm Chkl gene were positively expressed in all of the groups ;2)The expression of Chkl mRNA and protein in oligozoospermia,as- thenospermia and oligoasthenozoospermia were significantly lower than that in the normal(P〈0.05) , and the lowest level in the oligoasthenozoospermia.Conclusion The expression of sperm Chkl gene in infertility men in Han nation- ality living in middle altitude is considered down-regulated, suggesting that it may be the reason for effecting on se- men quality with the altitude hypoxia environment.
