摘 要:目的通过丙硫氧嘧啶(PTU)造成大鼠甲状腺肿的模型,观察贝牡莪消颗粒对大鼠甲状腺肿组织形态的影响,进而探究该药物对本病的治疗作用。方法选取40只雄性SD大鼠,随机选取其中10只作为空白组,其余30只给予丙硫氧嘧啶0.15 mg/g,1次/d,持续灌胃40 d。造模成功后,将30只大鼠随机分为3组,即模型对照组(2 m L/d)、贝牡莪消组(22.2 mg/g)、优甲乐组(0.08μg/g)。30 d后处死大鼠,光镜下观察大鼠甲状腺的组织形态。结果肉眼观察:PTU组大鼠甲状腺较空白组体积明显增大;镜下观察:PTU组与空白组对比,甲状腺滤泡间的差异明显,大小不一,呈圆形或椭圆形,滤泡腔变窄,呈现不规则的形状,腔内胶质数量明显降低。显微镜下观察大鼠甲状腺HE染色示:贝牡莪消组甲状腺滤泡与上皮细胞恢复较好,形态较为规则,基本恢复正常;而优甲乐组仍然可见到少许浸润的炎性细胞存在于滤泡腔和间质。结论 PTU能够导致大鼠甲状腺肿大;贝牡莪消组甲状腺细胞恢复程度明显优于优甲乐组,并能使大鼠甲状腺肿组织形态得到改善。Objective To investigate the effect of shellfish granule on the morphology of goiter in rats by probiotics(PTU), and to explore the therapeutic effect of the drug on the disease. Methods Forty male SD rats were randomly selected, and 10 of them were randomly selected as the blank group. The other 30 rats were given 0.15 mg/g of propylthiouracil and once/d for 40 days. Thirty rats were randomly divided into three groups: model control group(2 mL/d), shellfish(22.2 mg/g) and excellent group(0.08 μg/g). Rats were sacrificed 30 days later and the histomorphology of the thyroid was observed under light microscope. Results The naked eye: PTU group significantly increased the volume of thyroid gland compared with the blank group; Microscopic view: PTU group compared with the blank group, the difference between the thyroid follicles were significant, and sizes of them were different, round or oval, follicles cavity narrowed, showing irregular shape, the number of intracavity gel decreased significantly.Under the microscope, HE staining showed that the thyroid follicle and epithelial cells recovered from the shellfish were better than those in the control group. The morphological changes were normal and the control group showed that the infiltration of inflammatory cells was still cavity and interstitial. Conclusion PTU can lead to thyroid enlargement in rats. The recovery of thyroid cells in the shellfish was significantly better than that in the excellent group, whichcould improve the thyroid tissue morphology and treatment was safe and effective.
