摘 要:目的评价右美托咪定对糖尿病大鼠肺组织的保护作用。方法 40只成年雄性SD大鼠随机分为4组(n=10):正常组(N组)、不同剂量右美托咪定处理组(DEX1组、DEX2组)、糖尿病组(DM组)。DM组和DEX1组、DEX2组采用腹腔注射链脲佐菌素60mg/kg的方法制备糖尿病模型,N组仅腹腔注射等容量枸橼酸缓冲液。模型建立8周后,DEX1组和DEX2组每日在腹腔内分别注射右美托咪定5μg/kg、10μg/kg,N组和DM组每日仅腹腔注射等容量生理盐水。1周后,各组大鼠10%水合氯醛腹腔注射麻醉,经股动脉插管抽取动脉血作血气分析,随后处死,取肺组织,测定湿/干重比(W/D比)、光学显微镜观察并进行病理学评分、用ELISA法检测肺泡灌洗液(BALF)中TNF-α、IL-6的含量。结果与N组比较,DM组、DEX1组和DEX2组的PaO2显著下降(P<0.05),肺组织病理评分、TNF-α、IL-6含量显著升高(P<0.05)。DEX1组和DEX2组与DM组比较,大鼠动脉血PaO2明显提高(P<0.05),W/D比值、肺组织病理评分、TNF-α、IL-6含量显著下降(P<0.05)。结论右美托咪定可抑制糖尿病大鼠肺组织炎性反应,减轻糖尿病大鼠肺损伤。Objective To evaluate the protective effect of dexmedetomidine on lung injury in rats with streptozoto-cin-induced diabetes.Methods Fourty SD rats were randomly divided into 4 groups(n=10):normal group (group N), diabetes group (group DM),dexmedetomidine 1 group(group DEX1)and dexmedetomidine 2 group(group DEX2).DM rats model was induced by intraperitoneal streptozotocin 60 mg/kg and confirmed by blood glucose level〉16.7 mmol/L in groups DM,group DEX1and group DEX2.Citric acid buffer solution was intraperitoneal injected in group N.At 8 weeks after establishment of the Diabetic rats model,dexmedetomidine was intraperitoneal injected in daily respectively 5μg/kg,10 μg/kg in DEX1 and DEX2 group.Equal volume of normal saline was intraperitoneal injected in group N and DM.After 1 week,femoral artery intubation is drawn through arterial blood for blood gas analysis.Then the rats were sacrificed,the lungs were removed for determination of W/D lung weight ratio,examination of the pathological changes and grade.Additionally,we examined the concentration of tumour necrosis factor-alpha(TNF-α)and interleukin-6 (IL-6)in bronchoalveolar lavage fluid(BALF)by enzyme-linked immunosorbent assay.Results Compared with group N, PaO2 were significantly decreased in group DM,DEX1 and DEX2,the pathological grade,contents of TNF-α,IL-6 were significantly increased in group DM,DEX1 and DEX2(P 〈0.05).Compared with group DM,PaO2 were significantly decreased,W/D lung weight ratio,the pathological grade,contents of TNF-α,IL-6 were significantly decreased in DEX1and DEX2 group(P 〈0.05).Conclusion Dexmedetomidine can reduce lung injury in diabetic rats through inhibit inflammatory reaction.
