摘  要:目的:通过小干扰RNA(siRNA)下调Cyclin A1的表达,观察其对骨髓增生异常综合征(MDS)细胞系SKM-1细胞增殖的影响并探讨其作用机制。方法:采用阳离子脂质体转染方法,在SKM-1细胞中转染Cyclin A1的siRNA;转染48 h后收集细胞,应用Western blot及RT-PCR测定转染效率;应用CCK-8方法检测细胞增殖,Western blot及RT-PCR检测Cyclin A1基因沉默前后CDK2、RUNX1、SRSF2基因表达的水平。结果:转染Cyclin A1特异性siRNA 48 h后,Cyclin A1蛋白及mRNA表达水平显著下降(P<0.01)。下调Cyclin A1后,SKM-1细胞的增殖受抑,CDK2、RUNX1及SRSF2基因的蛋白及mRNA表达量均下调(P<0.05)。结论:Cyclin A1表达下调后,SKM-1细胞的增殖受到抑制,表明Cyclin A1可能是MDS治疗的潜在靶点之一。Objective: To investigate the effects of Cyclin A1 on the proliferation of SKM-1 cells and its underlying role in myelodysplastic syndrome( MDS). Methods: Cyclin A1 was knocked down with its small interfering RNA( siRNA).The efficiency of siRNA transfection was measured by Western blot and RT-PCR. Then the proliferation of SKM-1 cells and the expression of CDK2,RUNX1 and SRSF2 with and without knockdown of Cyclin A1 recorded and analysed respectively.Results: Cyclin A1 was knocked down by siRNA after transfected for 48 h. The kncokdown of Cyclin A1 inhibited the proliferation of SKM-1 cells and down-regulated the expression of CDK2,RUNX1 and SRSF2,and these effects were at least partially mediated through RUNX1 and SRSF2 signaling pathway. Conclusion: Cyclin A1 plays an important role in the proliferation of SKM-1 cells. These findings provide new insights into the pathogenesis of MDS,and it may be a potential target in the treatment of MDS.
