摘 要:旨在研究丙二酰CoA对小鼠脂肪细胞甘油三酯及其代谢产物的影响,将小鼠3T3-L1前脂细胞诱导成成熟的脂肪细胞做为研究对象,在诱导培养第14天,加入含0(对照组)、20μmol/L(试验Ⅰ组)、50μmol/L(试验Ⅱ组)、200μmol/L(试验Ⅲ组)的丙二酰CoA完全培养基培养细胞0、36、72h,收集细胞进行ELISA及后续检测。试验Ⅰ、Ⅱ、Ⅲ组合成的甘油三酯量高于对照组水平(P<0.01);试验各组乙酰CoA羧化酶、脂肪酸合成酶、柠檬酸等含量均高出对照组水平(P<0.01);试验各组激素敏感脂肪酶、肉碱脂酰转移酶Ⅰ、游离脂肪酸及乙酰CoA等含量均极显著低于对照组水平(P<0.01)。丙二酰CoA能够促进小鼠脂肪细胞甘油三酯的合成和沉积,且丙二酰CoA的浓度在一定范围内随着时间和甘油三酯合成量成正相关;50μmol/L丙二酰CoA为最佳浓度,在此浓度下脂肪细胞能够最大化的合成甘油三酯;丙二酰CoA还能提高甘油三酯合成代谢中乙酰CoA羧化酶、脂肪酸合成酶、柠檬酸等产物的含量(P<0.01);能够降低甘油三酯分解代谢中激素敏感脂肪酶、肉碱脂酰转移酶Ⅰ、游离脂肪酸及乙酰CoA等产物的含量(P<0.01)。The purpose of this study is to investigate the effect of malonyl CoA on triglyceride and its metabolites in mouse adipocytes.Firstly,the mouse 3 T3-L1 cell was induced into mature adipocytes as the research subject,the cell were cultured in malonyl CoA complete medium containing 0μmol/L(CK group),20μmol/L(groupⅠ),50μmol/L(groupⅡ)and 200μmol/L(group Ⅲ)on the 14 th day,and the cells were collected at 0 h,36 hand 72 hfor the detection of ELISA and other substances.The amount of TG in the experimental group Ⅰ,Ⅱ and Ⅲ was significantly higher than that of the control group(P <0.01).The levels of ACC,FAS and citrate in the test groups were significantly higher than those of the control group(P <0.01).The levels of HSL,CPT1,FFA and acetyl CoA in the test groups were significantly lower than those of the control group(P <0.01).The malonyl CoA could promote the synthesis and deposition of TG in mouse adipocytes,and the concentration of malonyl CoA was positively correlated with the amount of TG.50μmol/L malonyl CoA was the best concentration.In this level the mouse adipocytes could maximize the synthesis of TG.The molonyl CoA could also significantly increase the content of ACC,FAS,citrate and other products in TG synthesis and reduce the content of HSL,CPT1,FFA,acetyl CoA and other products in TG disintegration(P <0.01).
