两波长变化HPLC法同时测定胆胰康泰散中4种成分

摘　　要：目的利用高效液相色谱法,建立胆胰康泰散中芍药苷、黄芩苷、川楝素、柴胡皂苷d的含量测定方法。方法采用HPLC法,色谱柱选用Inertsil C18柱(250 mm×4.6 mm,5μm),柱温选用35℃,用乙腈-体积分数0.1%磷酸水溶液为流动相,采取梯度洗脱,流速设为1.0 mL·min^(-1),检测波长选用230 nm、210 nm。结果芍药苷、黄芩苷、川楝素和柴胡皂苷d分别在质量浓度8.75~280.00 mg·L^(-1)(r=0.999 9)、12.50~400.00 mg·L^(-1)(r=0.999 1)、232.50~7 440.00 mg·L^(-1)(r=0.999 8)和21.25~680.00 mg·L^(-1)(r=0.999 5)内与峰面积呈良好线性关系;回收率分别为102.9%(RSD=0.5%)、100.2%(RSD=0.8%)、102.1%(RSD=0.7%)和98.5%(RSD=1.1%)。结论本方法简便、可靠、重现性好,适用于研究胆胰康泰散中有效成分的含量测定。Objective To establish an HPLC method for simultaneous determination of paeoniflorin, baicalin, azadirachtin,and saikosaponin D in Danyikangtai powder. Methods HPLC was performed on a Inertsil C18 column （250 mm× 4. 6 mm, 5 μm） with gradient elution of acetonilrile and 0. 1% phosphoric acid at the flow rate of 1.0 mL .min-1, detection wavelength at 230 rim,210 nm and column temperatureat 35 ℃. Results The calibration curve was linear within the range of 8.75 -280. 00 mg .L-1（ r = 0. 999 9）, 12. 50 - 400. 00 mg.L -1 （ r = 0. 999 1 ）, 232. 50 - 7 440. 00 mg- L -1 （ r = 0. 999 8 ） and 21.25 - 680. 00 mg. L-1 （ r = 0. 999 5 ） for paeoniflorin, baicalin, azadirachtin and saikosaponin D, respectively. Conclusions The method is simple, accurate and reproducible, which can be used for the quality control of Danyikangtai powder.
