银屑病发病诊断及治疗中的MPAK/NF-kB信号通路作用

摘　　要：目的探讨MPAK/NF-κB信号通路在银屑病发病、诊断及治疗中的表达和意义。方法选取2015年5月-2017年5月本院收集的90例中度和重度寻常型银屑病患者,给予口服阿维A治疗8周,采用免疫组织化学法和Western blot法检测银屑病患者皮肤组织中MAPK和NF-κB蛋白的表达;ELISA法检测患者血清中MAPK、NF-κB、TNF-α、IL-6、IL-8含量。结果与对照组相比,进行期和稳定期患者血清内MAPK、NF-κB、TNF-α、IL-6和IL-8含量显著升高;MAPK和NF-κB蛋白含量显著增加,差异具有统计学意义(P<0.05);与进行期相比,稳定期患者血清内MAPK、NF-κB、TNF-α、IL-6和IL-8含量显著降低,患者体内MAPK和NF-κB蛋白含量明显降低,差异具有统计学意义(P<0.05);与治疗前相比,在阿维A治疗后,进行期和稳定期血清内MAPK、NF-κB、TNF-α、IL-6和IL-8含量显著降低,患者体内MAPK和NF-κB蛋白含量均明显下降,差异具有统计学意义(P<0.05)。结论 MPAK/NF-κB信号通路在银屑病发病、诊断及治疗中具有重要的作用;阿维A对银屑病的治疗具有一定的临床意义。Objective To investigate the expression and significance of MPAK/NF-KB signaling pathway in the pathogene- sis, diagnosis and treatment of psoriasis. Methods From May 2015 to May 2017, 90 patients with moderate and severe psoriasis were collected and treated with oral administration of Etretin for 8 weeks. The expression of MAPK and NF- KB protein in the skin tissues of patients with psoriasis were detected by immunohistochemistry and Western blot. The levels of MAPK, NF-KB, tumor necrosis factor alpha （ TNF- α ）, interleukin-6 （ IL-6 ） and interleukin-8 （ IL-8 ） content. Results Compared with the control group, the levels of MAPK, NF-KB, TNF-α, IL-6 and IL-8 in serum were significantly increased in the patients with stable phase and stable stage, MAPK and NF-KB protein content increased significantly, the difference was statistically significant （P〈 0.05）. The contents of MAPK, NF-KB, TNF-α, IL-6 and IL-8 in the serum of patients with stable phase were significantly lower than those in the control group, the content of MAPK and NF-KB protein in patients was significantly decreased, the difference was statistically significant （P〈0.05）. Compared with before treatment, after treatment with acitretin, the levels of MAPK, NF-KB, TNF-α, IL-6 and IL-8 in serum and serum were significantly decreased, the contents of MAPK and NF-KB in the patients were significantly decreased, the difference was statistically significant （P〈O.05）. Conclusion MPAK / NF-KB signaling pathway plays an important role in the pathogenesis, diagnosis and treatment of psoriasis. Etretin has a certain clinical significance in the treatment of psoriasis.
