摘  要:目的探讨健脾消癌方拮抗结直肠癌转移的可能作用机制。方法 60只BALB/c-nu裸鼠随机分为模型组、空白组及中药低、高剂量组。除空白组外,其余裸鼠采用盲肠黏膜内注射HCT116细胞方法建立人结直肠癌裸鼠移植模型。中药低、高剂量组分别予健脾消癌方10、40 g/(kg·d)灌胃;模型组和空白组以生理盐水2 ml/100 g灌胃。每日1次,连续给药4周。运用Western Blot方法检测肿瘤组织中E-钙粘蛋白、波形蛋白、转化生长因子β(TGF-β)蛋白表达量;比较各组大鼠结肠癌肝、肺、淋巴转移情况。结果中药高、低剂量组肝、肺、淋巴转移率低于模型组(P<0.05),中药高剂量组的肝、肺转移率低于低剂量组(P<0.05)。与空白组比较,各组E-钙粘蛋白表达均明显降低,波形蛋白、TGF-β表达均明显升高(P<0.05或P<0.01);与模型组比较,中药高、低剂量组E-钙粘蛋白表达均明显升高,波形蛋白、TGF-β表达均明显降低(P<0.05或P<0.01),中药高、低剂量组各指标比较差异无统计学意义(P>0.05)。结论健脾消癌方通过TGF-β介导而上调E-钙粘蛋白、下调波形蛋白表达,逆转人结直肠癌裸鼠移植模型肿瘤组织上皮间质转化进程,可能是其拮抗结直肠癌转移的作用机制之一。Objective To explore the potential mechanism of Jianpi Xiao'ai Fang (Spleen-fortifying Cancer- dissolving Decoction, 健脾消癌方, JXF) in antagonizing the metastasis of colorectal cancer. Methods Sixty BALB/ c-nu nude mice were randomized into a model group, a blank group and Chinese medicine low and high dose groups. The mice in each group were given HCT116 intramucosal injection into the cecum to establish a human colorectal cancer xenograft model except for those in the blank group. Chinese medicine low and high dose groups were respec- tively given JXF at the doses of 10 or 40 g/( kg· d) by garage; the model group and the blank group were given 2 ml/ 100 g of normal saline by gavage. The interventions were all given once a day for 4 weeks. The expressions of E- cadherin, vimentin, and transforming growth factor-3 (TGF-β) in the tumor tissue were detected by Western Blot; the liver, lung and lymphatic metastases of colon cancer in each group were compared. Results The liver, lung and lymphatic metastasis rates in Chinese medicine high and low dose groups were lower than those in the model group (P 〈 0.05 ). The rates of liver and lung metastasis in Chinese medicine high dose group were lower than those in the low dose group ( P 〈 0.05 ). Compared with those in the blank group, the expression of E-cadherin significantly decreased, while the expressions of vimentin and TGF-β significantly increased in other groups ( P 〈 0. 05 or P 〈 0. 01); compared with those in the model group, the expressions of E-cadherin significantly increased, while the expressions of vimentin and TGF-~ significantly decreased in Chinese medicine high and low dose groups (P 〈 0. 05 or P 〈 0.01 ). There was no significant difference in alhhe indexes between the high and low dose groups (P 〉 0.05 ).
