摘 要:目的观察健脑益寿胶囊(JYC)对阿尔茨海默病(AD)小鼠海马RAGE、LPR1、ApoE的影响,初步探讨JYC是否通过影响BMEC功能发挥神经元保护作用,探究药物的作用靶点。方法 APP/PS1双转基因小鼠(3月龄),聚合酶链(PCR)法验证APP和PS1基因表达。APP/PS1小鼠随机分4组:模型组、阳性药对照组(多奈哌齐)、JYC高、低剂量组、空白对照组(C57BL/6)。模型组和空白对照组给生理盐水,各组动物连续灌胃4周,采用免疫组织化学法(IHC)、光学显微镜下观察小鼠海马RAGE、LRP1、ApoE蛋白表达。结果光镜下观察结果显示,JYC与模型组比较,高、低剂量组海马LRP1和ApoE表达增加,RAGE表达降低。结论研究表明JYC高、低剂量可改善AD小鼠学习记忆能力,其机制可能与JYC增强AD小鼠海马RAGE蛋白表达,降低AD小鼠海马LRP蛋白表达相关,进而改善Aβ诱导的脑微血管内皮细胞损伤,对脑微血管内皮细胞损伤产生保护作用。Objective To investigate the effect of Jian - nao - yi - shou capsule (JYC) on the expression of RAGE, LPR1, ApoE in hippocampus of Alzheimer's disease (AD) mice, to explore whether JYC can play the role of neuron protection by influencing the function of BMEC, and to explore the targets of drug. Methods App/PS1 double transgenic mice (3 months old), the expression of App and PSI were confirmed by polymerase chain reaction (PCR). App/PS1 mice were randomly divided into 4 groups : model group, positive drug control group ( donepezil), jyc high - dose group, jyc low - dose group, blank control group. The model group and the blank control group were given saline, each group of animals were given oral administration continuously for four weeks, the protein expression of RAGE, P1 and ApoE in hippocampus of mice was observed by immunohistochemistry (IHC) and optical microscope. Resuits The results of light microscope showed that the expression of P1 and ApoE in the bippocampus of high and low dose groups increased, however, the expression of RAGE decreased compared with the model group. Conclusion The study showed that high and low dose of JYC can improve the learning and memory ability of AD mice, The mechanism may he related to JYC increase the expression of RAGE protein , and decrease the expression of LRP protein in hippocampus of AD mice, and improve the injury of brain microvascular endothelial cells induced by Aβ, and play protect effect on the brain microvascular endothelial cells injury'.
