摘  要:目的:对肥胖大鼠进行四周有氧运动和节食双重干预,观察大鼠体重和下丘脑Ghrelin的变化情况,来探究运动结合节食对下丘脑Ghrelin的影响以及Ghrelin在减肥中可能起到的作用。方法:用220只SD大鼠建立肥胖模型后,将肥胖大鼠随机分为肥胖对照组(OC)、运动节食组(ED)、运动节食即刻组(ED1)和运动节食3h组(ED2)。ED组进行游泳运动4周,每周6天,每天60分钟。建模期间和大鼠运动期间,每周测量大鼠体重,第四周运动结束后即刻和3小时取材,进行免疫组织化学实验并观察下丘脑Ghrelin的阳性表达。结果:ED组的大鼠在运动期间体重下降明显,在第一周末与NC组相比有显著性差异,在第四周末显著性降低;OC组、ED1组、ED2组大鼠的下丘脑Ghrelin的阳性细胞数量都显著减少(P<0.05),运动即刻后下丘脑Ghrelin表达增加,运动3h后下降。结论:有氧运动结合控食能够有效减轻大鼠体重,并且降低大鼠下丘脑Ghrelin的表达。Objective: The obese rats were given a four-week intervention with aerobic exercise and diet,and then observe the changes of body weight and hypothalamic Ghrelin to explore the effect of exercise combined with dieting on Ghrelin in the hypothalamus and the possible role of Ghrelin in weight loss. Methods: General obesity model is set up with 220 SD rats,the obesity rats were randomly divided into obese control group( OC),exercise diet group( ED),exercise diet instantly group( ED1) and exercise diet 3 h group( ED2). ED group for four weeks of swimming,6 days a week and 60 minutes a day. During the modeling period and swimming,measure the body weight weekly,immediately after exercise and 3 hours after exercise to take the organization at the end of forth week,immunohistochemistry was then performed to observe Ghrelin in the hypothalamus. Results: the weight of the rats in the ED group was significantly lower during swimming,there was a significant difference between the ED group and the NC group at the end of first week,and significant reduction in the end of fourth week; the number of positive cells of Ghrelin in the hypothalamus of the OC group,ED1 group and ED2 group was significantly reduced( P < 0. 05),the expression of Ghrelin in the hypothalamus was increased immediately after exercise and decreased after 3 h. Conclusions: The combination of aerobic exercise and controlled diet can effectively reduce the weight of rats and reduce the expression of Ghrelin in the hypothalamus of rats.
