摘  要:目的研究商陆皂苷甲的肠黏膜损伤与泻下作用及其机制。方法实验分为空白组、商陆生品组、商陆皂苷甲高剂量组和低剂量组。免疫组织化学和实时荧光定量PCR的方法检测各实验组大鼠结肠黏膜粘蛋白2(MUC2)和水通道蛋白9(AQP9)的表达。结果 PCR结果显示商陆生品组、商陆皂苷甲高剂量组和低剂量组MUC2表达较空白组降低(P<0.05)。商陆生品组、商陆皂苷甲高剂量组和低剂量组AQP9的表达较空白组升高(P<0.05)。免疫组化结果显示空白组MUC2阳性明显,着棕黄色。商陆皂苷甲低剂量组可见零星的棕黄色阳性着色,商陆生品组和商陆皂苷甲高剂量组未见棕黄色。空白组AQP9着色浅,商陆生品组、商陆皂苷甲高剂量组和低剂量组阳性着色加深。结论商陆皂苷甲和商陆生品均可以降低大鼠结肠黏膜组织MUC2的表达损伤结肠黏膜。通过增加肠黏膜AQP9的表达发挥泻下作用。Objective To discuss the effects of esculentoside A on intestinal mucosal damage and its purgative function. Methods In the experiment,a blank group,a raw material of radix phytolaccae group( raw material group),a high-dose esculentoside A group( high-dose group) and a low-dose esculentoside A group( low-dose group) were divided. The IHC and RT-PCR were adopted to determine the expressions of MUC2 and AQP9 in the colonic mucosa in the rats. Results It is shown in PCR that the MUC2 expressions of the raw material group,the high-dose group and the low-dose group were lower than the blank group( P〈0. 05). The expressions of AQP2 in the raw material group,the high-dose group and the low-dose group were higher than the blank group( P〈0. 05). IHC test showed that MUC2 was remarkably positive,stained as brown yellow in color in the blank group. It was slightly positive-stained in the low-dose group. The brown yellow color of it was not observed in the raw material group and high-dose group. AQP9 was less colored in the blank group and it was deeply colored in the high-dose and low-dose groups. Conclusion Esculentoside A and the raw material of radix phytolaccae reduces MUC2 expression in colonic mucosa to damage the mucosa. It achieves the purgative role through increasing AQP9 expression in intestinal mucosa.
