摘 要:目的:制备贝伐珠单抗靶向载紫杉醇超顺磁性氧化铁纳米粒,以期靶向治疗肿瘤。方法:热分解法制备超顺磁性氧化铁纳米粒,电镜观察形态、粒径,利用振动样品磁强计分析其饱和磁化强度;利用旋转蒸发法制备载紫杉醇超顺磁性氧化铁纳米粒,电镜下观察形态、粒径,并计算其包封率以及载药量;以贝伐珠单抗和载紫杉醇超顺磁性氧化铁纳米粒为原料合成Bevacizumab靶向载紫杉醇超顺磁性氧化铁纳米粒,电镜下观察其形态特征。结果:(1)超顺磁性氧化铁纳米粒外观为黑褐色,分布均匀;电镜下观察纳米粒呈等轴晶形,平均粒径大小约为9.5 nm,其饱和磁化强度为53.32 emu·g^(-1)。(2)载紫杉醇超顺磁性氧化铁纳米粒的粒径约为18.89 nm,有效载药量为0.2 mg·(mg Fe)^(-1),包封率约为(97.34±1.58)%。(3)贝伐珠单抗靶向载紫杉醇超顺磁性氧化铁纳米粒的形态特征结果显示,纳米粒粒径120~200 nm,粒径大小均匀,部分呈融合。结论:通过热分解法、旋转蒸发法等合成的贝伐珠靶向载紫杉醇超顺磁性氧化铁纳米粒具有良好的载药特性。Objective:To prepare bevacizumab targeted paclitaxel loaded superparamagnetic iron oxide nanoparticles.Methods:Superparamagnetic iron oxide nanoparticles were prepared by thermal decomposition synthesis,and the magnetization saturation was detected by vibrating magnetometer.Using rotary evaporation method,paclitaxel loaded superparamagnetic nanoparticles were prepared,whose morphology,particle size,the encapsulation rate and drug loading were observed under electron microscopy.Bevacizumab targeting paclitaxel loaded superparamagnetic iron oxide nanoparticles were synthesized with bevacizumab monoclonal antibody and paclitaxel loaded superparamagnetic iron oxide nanoparticles.Results:Their morphology distribution was observed under the electron microscope:nanoparticles were crystal,with an average size of 9.5 nm,the saturation magnetization of 53.32 emu·g^-1;the size of paclitaxel(PTX) with superparamagnetic iron oxide nanoparticles was 18.89 nm,effective drug loading was 0.2 mg·(mg Fe)^-1.The encapsulation rate was about(97.34±1.58) %.The characteristics of paclitaxel loaded nanoparticles with superparamagnetic iron oxide nanoparticles targeted at monoclonal antibody bevacizumab showed that the particle size was about 120-200 nm.Conclusion:Bevacizumab targeted paclitaxel loaded superparamagnetic iron oxide nanoparticles synthesized by thermal decomposition method and rotating evaporation method are easily loaded.
