聚肌胞对禽流感油乳剂疫苗的体液免疫增强作用研究

摘　　要：为了研究聚肌胞是否能够增强油乳剂灭活疫苗免疫后鸡群的体液免疫应答,将3个不同来源的聚肌胞产品与H5亚型禽流感病毒灭活抗原相混合,使用轻质矿物油乳化制备疫苗,分别免疫21日龄SPF鸡,同时设置添加抗菌肽的疫苗免疫组、常规油乳剂疫苗免疫组以及不免疫对照组。在免疫后第14、21、28、35 d分别翅下静脉采集SPF鸡抗凝血,通过血凝抑制试验检测鸡血清中禽流感抗体的水平,使用统计学方法评价疫苗中添加聚肌胞对鸡群体液免疫反应的影响。结果发现,添加了聚肌胞或抗菌肽的疫苗免疫后,鸡血清中禽流感抗体水平显著高于普通油苗组。另外,不同来源的聚肌胞产品(PIC-1、PIC-2和PIC-3)免疫增强作用存在差异,以PIC-1免疫增强效果最好。在PIC-1疫苗免疫后,鸡血清抗体增长快,且可维持较高水平。总之,本研究表明聚肌胞是良好的禽病毒性疫苗免疫增强剂;不同厂家的聚肌胞产品质量有差异,应择优使用。To evaluate the efficacy of poly I∶ C( PIC) as adjuvant for avian influenza vaccines,three different sources of PIC were mixed with viral antigens of H5 avian influenza virus,and oil emulsion vaccines were developed. Twenty-one-day-old specific-pathogen-free( SPF) chickens were vaccinated with either vaccine. Meanwhile,an unvaccinated chicken group,a cecropin-added oil emulsion vaccine group and a common oil emulsion vaccine( vaccine control) group were set. Chicken blood was collected on the 14 th,21 st,28 th and 35 th day after vaccination to test serum antibody levels against avian influenza by hemagglutination inhibition assay. Statistic analysis showed that chickens vaccinated with PICs or cecropin adjuvant developed significantly higher antibody level compared to the vaccine control group. Further,PIC of different sources( PIC-1,PIC-2 and PIC-3) showed varied immune-enhancing effects and that of PIC-1 wasthe best. After vaccinated with PIC-1,the serum antibody of chickens increased fast and could maintain at higher level. In conclusion,PIC is a better promising adjuvant for chicken viral vaccine,and its efficacy may vary due to the sources.
