摘 要:目的分析麝香通心滴丸对心肌梗死小鼠微循环障碍的改善作用并探讨其相关机制。方法选用6周~8周C57雄性小鼠30只,随机分为假手术对照组、微循环障碍模型组[心肌梗死手术合并脂多糖诱导(MI+LPS)组]、模型联合麝香通心滴丸干预(SXTXDW)组。实验终点处死动物,检测内皮型一氧化氮合酶(e NOS)、神经型一氧化氮合酶(n NOS)在提睾肌的表达。结果与假手术对照组比较,MI+LPS组微循环血流速度明显降低(P<0.05),微循环中白细胞黏附数目明显增多(P<0.05)。与MI+LPS组比较,SXTXDW组微循环血流速度明显增快(P<0.05),白细胞黏附数量明显减少(P<0.05),且提睾肌e NOS和n NOS表达明显增强(P<0.05)。结论麝香通心滴丸可改善心肌梗死小鼠微循环障碍,其机制可能与内源性一氧化氮产生增加、白细胞黏附减少等有关。Objective To explore the protective effects of Shexiang Tongxin dropping pill ( STDP)on microcirculation dysfunction inmice and its mechanism.Methods Thirty male C57 mice were used, of which were randomly divided into sham control group, myo -cardial infarction ( MI) +lipopolysaccharides ( LPS)group, and STDP group.The sham control group mice were given sham operation.And the other two groups were given myocardial infarct operation.After one week of operation, sham control group mice were intrap -eritoneally injected with NS, while the other two groups were injected with LPS.After 30min, the microcirculation flow velocity of cre -master was observed.Then the different means of intervention were given to the three group mice.The microcirculation flow velocity atdifferent time points and white blood cells adhesion were recorded.At last, the relative expression of endothelial nitric oxide synthase( eNOS)and neuronal nitric oxide synthase ( nNOS)mRNA were measured.Results Compared with those of sham control group,the microcirculation flow velocity of MI+LPS group was decreased significantly ( P 〈0.05) and the white blood cells adhered to en -dothelial cells of MI+LPS group was increased ( P 〈0.05) .Compared with those of MI+LPS group, the microcirculation flow velocityof STDP group increased significantly ( P 〈0.05) and the white blood cells adhered to endothelial cells of STDP group decreased ( P 〈0.05) , the relative expression of eNOS and nNOS mRNA of STDP group increased significantly ( P 〈0.05) .Conclusion STDP canimprove the microcirculation in mice.The more produced endogenous NO and the decreased white blood cells adhered to endothelialcells might be the mechanism.
