通心络干预肥大细胞脱颗粒诱导的炎症反应对缺血再灌注心肌的保护机制

摘　　要：目的探讨通心络干预肥大细胞脱颗粒诱导的炎症反应对缺血再灌注模型大鼠心肌损伤的保护机制。方法将36只健康成年雄性SD大鼠随机分成假手术组、模型组和通心络组。通过可逆性左冠状动脉前降支结扎法建立心肌缺血再灌注模型,大鼠心肌缺血1 h,再灌注2 h;假手术组仅穿线不结扎。通心络组大鼠予通心络灌胃,假手术组和模型组给予等量的生理盐水灌胃。1周后,免疫组化法检测心肌标本类胰蛋白酶(TPS)、c-Fos和组胺(Histamine);Elisa法检测血清TPS和c-Fos及TUNEL凋亡指数等指标。结果通心络组大鼠的心肌凋亡指数和TPS、c-Fos、组胺表达水平较模型组均明显降低(P<0.05或P<0.01)。结论通心络可抑制心肌细胞凋亡,并通过降低血清TPS和c-Fos及组胺表达水平,抑制肥大细胞脱颗粒诱导的炎症反应,保护受损心肌。Objective To investigate the effect of protective effect and mechanism of Tongxinluo capsule on myocardial injury inducedby mast cells in rats with ischemia reperfusion injury. Methods Thirty six healthy adult male Sprague Dawly （ SD）rats were ran -domly divided into sham operation group, model group, Tongxinluo group.Myocardial ischemia reperfusion model was established byligation of the left anterior descending coronary artery, rat myocardial ischemia 1 h, reperfusion 2 h; while the sham group was punc -tured without ligation.The rats in Tongxinluo group were intragastric administration by Tongxinluo suspension, and the model groupand the sham group were intragastric administration by saline.After 1 weeks, the expressions of tryptase, c Fos, and histamine weredetected by immunohistochemistry.ELISA method was used to detect serum TPS and c Fos, and TUNEL apoptosis index. Results The apoptosis index of the Tongxinluo group rats, the expressions of Tryptase, c Fos, and histamine were significantly lower thanthose in the model group （ P 〈 0.01） .Conclusion Tongxinluo capsule can inhibit the apoptosis of myocardial cells, and by reducingTryptase, c Fos and histamine,Inhibition of inflammatory response induced by degranulation of mast cells, reduce myocardial inju -ry, thus protecting the myocardial tissue.
