摘 要:目的探讨生长分化因子11(GDF11)对T2DM小鼠胰岛β细胞的作用。方法选取C57BL/6J野生型小鼠,通过高脂饮食及STZ制备T2DM模型并随机均分为糖尿病rGDF11干预组(DM+rGDF11)和糖尿病对照组(DM),同时将普通饲料喂养的C57BL/6J小鼠随机分为正常rGDF11干预组(Con+rGDF11,n=10)和正常对照组(Con,n=10),观察GDF11对小鼠生理生化指标、胰岛β细胞功能及含量指标的影响。结果干预6周后,Con组与Con+rGDF11组各糖脂代谢指标比较,差异无统计学意义(P>0.05)。与DM组比较,DM+rGDF11组FBG、HbA1c、胰升血糖素及血脂水平降低(P<0.05),且rGDF11干预后上调胰岛β细胞重要基因表达,促进胰岛素分泌,改善胰岛形态及β细胞含量(均P<0.05)。结论 GDF11可改善胰岛β细胞功能及维持β细胞含量,进而延缓糖尿病的发生发展。Objective To investigate the effects of growth differentiation factor 11(GDF11)on pancreaticβcell in type 2 diabetic mouse model. Methods C57 BL/6 Jmice were rendered diabetic by feeding with high fat diet plus intraperitoneal injection of streptozotocin.And then,all the diabetic mice were divided into two groups randomly:DM+rGDF11 group and DM group.Normal control mice were randomly divided into Con+rGDF11 group(n=10)and Con group(n=10).The effects of GDF11 on metabolic parameters,βcell function and βcell mass were evaluated. Results After 6 weeks' intervention,the metabolic parameters were not significantly different in Con group and Con+rGDF11 group(P〈0.05).FBG,HbA1 c,serum glucagon and lipid profiles were decreased in DM+rGDF11 group than in DM group(P〈0.05).rGDF11 administration upregulated expression ofβcell-specific genes,improved insulin secretory response,improved islet morphology andβcell mass(all P 0.05).Conclusion GDF11 may preserve isletβcell function and mass and thus may delay the progression of diabetes.
