摘 要:建立一种从萝卜芽苗中提取、纯化还原型萝卜硫苷(4-甲硫基-3-丁烯基硫代葡萄糖苷)的方法。采用醇提-水沉法从萝卜芽苗中提取还原型萝卜硫苷,提取物中还原型萝卜硫苷含量为11.56%;经酸性氧化铝柱层析预纯化得到还原型萝卜硫苷粗品,还原型萝卜硫苷含量为76.01%;经制备液相色谱进一步纯化得到还原型萝卜硫苷纯品,还原型萝卜硫苷含量为98.75%。采用质谱、核磁共振对还原型萝卜硫苷纯品进行结构鉴定,结果符合还原型萝卜硫苷的分子结构特征。醇提-水沉法可快速提取还原型萝卜硫苷,酸性氧化铝柱层析结合制备液相色谱可高效纯化还原型萝卜硫苷。To establish a method for the extraction and purification of glucoraphasatin(4-Methylsulfanyl-3-butenyl glucosinolate) from radish sprouts. Glucoraphasatin was extracted with alcohol extraction and water sedimentation method from radish sprouts, the content of glucoraphasatin in the extract was 11.56%. The obtained extract was pre-purified with acidic alumina based column chromatography to yield crude glucoraphasatin, and the content of glucoraphasatin was 76.01%. The crude glucoraphasatin was purified with preparative liquid chromatography to obtain pure glucoraphasatin, and the content of glucoraphasatin was 98.75%. The chemical structure of pure glucoraphasatin was identified by MS and NMR, the identification results were consistent with the molecular structure characteristics of glucoraphasatin. The alcohol extraction and water sedimentation method could rapidly extract glucoraphasatin from radish sprouts, and the acidic alumina based column chromatography combined with preparative liquid chromatography could efficiently purify glucoraphasatin.
