摘 要:目的明确骨关节炎患者血浆及关节液中脂联素亚型的分布特征。方法山西大医院骨科2015年11月至2016年5月因骨关节炎住院行人工全膝关节表面置换术的患者10例10膝,收集血浆及关节液标本,采用凝胶过滤层析、非变性聚丙烯酰胺凝胶电泳结合蛋白质印迹的方法,对血浆及关节液中脂联素亚型进行测定和对比。结果骨关节炎患者血浆及关节液经凝胶过滤层析后进行脂联素测定均出现4个峰,非变性凝胶电泳蛋白质印迹分析显示,前2个为高相对分子质量脂联素,第3个为中相对分子质量脂联素,最后1个为低相对分子质量脂联素;关节液的脂联素亚型各峰值均低于血浆中相应的脂联素亚型各峰值。结论骨关节炎患者关节液中脂联素亚型的分布特征与血浆类似,但各峰值均降低。Objective To determine the distribution pattern of adiponectin subtypes in plasma and synovial fluid of patients with osteoarthritis(OA). Methods Included in this study were 10 OA patients(10 knees) who underwent total knee arthroplasty in the Department of Orthopedics, Shanxi Dayi Hospital, between November 2015 and May 2016. The patients were collected for plasma and synovial fluid samples. Gel filtration chromatography, and non-denaturing polyacrylamide gel electrophoresis combined with Western blotting were used to determine and compare the adiponectin subtypes between plasma and synovial fluid. Results There were four peaks of adiponectin detected in plasma and synovial fluid samples from OA patients on gel filtration chromatography. Non-denaturing gel electrophoresis and Western blotting showed that the first two peaks were of high molecular weight adiponectin, the third was of mid-molecular weight adiponectin and the last one was of low molecular weight adiponectin. The peaks of syn-ovial adiponectin subtypes were lower than their corresponding peaks in plasma samples. Conclusion The distribution pattern of adiponectin subtypes in synovial fluid is similar to that in plasma among patients with OA, but shows lower peak values.
