阳虚质膝骨性关节炎关节软骨中PKC的表达研究

摘　　要：目的:研究阳虚质膝骨性关节炎(Knee Osteoarthritis,KOA)与蛋白激酶C(PKC)表达之间的相关性。方法:从病例调查数据库中随机选取KOA病例共20例,其中非阳虚质KOA患者10例,阳虚质KOA患者10例;非KOA健康志愿者10例作为对照组。采用免疫组化及RT-PCR方法检测PKC在不同组别中的表达。结果:PKC在正常关节软骨及阳虚质KOA与非阳虚质KOA中均有表达,表达主要位于细胞浆及细胞核内,组间进行两两比较,差异有统计学意义(P<0.05);PKC基因在对照组与KOA病例组关节软骨组织中的表达进行比较,差异有统计学意义(P<0.01);PKC基因在阳虚质病例组、非阳虚质病例组与对照组进行两两比较差异有统计学意义(P<0.05)。结论:PKC基因与KOA的发病有一定相关性,且可能与阳虚质高发KOA有一定联系。Objective：To study the relativity between knee osteoarthritis （KOA） of Yang deficiency pattern and PKC expressions.Methods：All 20 KOA cases were randomly chosen from case investigation database,among them,there were ten cases with KOA of non Yang deficiency pattern and ten cases with KOA of Yang deficiency pattem;and ten non-KOA healthy volunteers were taken as the control group.The expressions of PKC in different groups were detected by using immunohistochemical method and RT-PCR method.Results：There were the expressions of PKC in normal arthrodial cartilage,KOA of Yang deficiency pattern and KOA of non Yang deficiency pattern,it mainly expressed in cytoplasm and nucleus,the difference showed statistical meaning in the comparisons among the groups （P〈0.05）;the difference had statistical meaning in the comparisons of the expressions of PKC in arthrodial cartilage between the control group and KOA case group （P〈0.05）;the case group of Yang deficiency pattern and the case group of non Yang deficiency pattern were compared with the control group in the expressions ofPKC gene,and the differences had statistical meaning （P〈0.05）.Conclusion：PKC gene is related to the incidence of KOA,and it might be related to the high incidence of KOA of Yang deficiency pattern to a certain degree.
