摘  要:双组份信号传导系统(TCS)是细菌最常见的膜传导系统,在细菌对外界环境胁迫响应机制中发挥着的重要作用。贪铜菌属细菌是典型的重金属抗性细菌,对多种重金属具有抗性。但是,迄今为止,有关贪铜菌的TCS研究尚不多见。本研究基于前期研究结果,运用全基因组生物信息学分析的方法,以Cupriavidus gilardii CR3为模式菌株,鉴定和分析了Cupriavidus gilardii的双组份信号传导系统基因分布及编码蛋白结构特征。结果表明,Cupriavidus gilardii CR3共有96个双组份信号系统基因。其中,有22个组氨酸激酶基因和反应调节因子成对存在组成双组份信号传导系统,12个融合组氨酸激酶,13个孤儿组氨酸激酶和27个孤儿反应调节因子。与已有文献报道的Cupriavidus metallidurans CH34、Cupriavidus necator N-1和Cupriavidus taiwanensis LMG 19424的双组分信号传导系统相比,Cupriavidus gilardii CR3的双组份信号系统较少,而融合组氨酸激酶和孤儿反应调节因子所占比例较大。Cupriavidus gilardii CR3含有特有组氨酸激酶功能域(PDB,MEDS,Kdp D)和反应调节因子功能域(ABC),这与Cupriavidus gilardii CR3的生存环境密切相关。本研究有助于全面了解贪铜菌属细菌双组分信号传导系统的功能机制,挖掘其在生物领域的应用潜力。Two-component systems (TCS) are commonly used for transmembrane signal transduction in bacteria, They play an important role in the adaptation mechanisms between bacteria and the environment. The genus Cupriavidus is a typical heavy metal-resistant bacterium, having resistance to various heavy metals. However, there were few studies of TCS in Cupriavidus yet. Based on the results of previous researches and the method of genome-wide bioinforrnatics analysis, this study used Cupriavidus gilardii CR3 as type strain, identified and analyzed the gene of TCS in Cupriavidus gilardii as well as its architectural feature of encoded protein. The result showed that CR3 totally contained 96 TCS genes, including 22 TCS clusters (formed by 22 couples of ilK genes and response regulators), 12 hybrid HKs, 13 orphan HKs, and 27 orphan RRs. /n comparison to the TCS of Cupriavidus metallidurans CH34, Cupriavidus neeator N-1 and Cupriavidus taiwanensis LMG 19424 in existing literature reports, C. gilardii CR3 possesses fewer TCS but more paired HKs and orphan RRs. C. gilardii contains unique HK (PDB, MEDS, KdpD) and RR (ABC) function domains. This situation closely related with the habitat of C. gilardii CR3. This research will be helpful for the comprehensive understanding of the versatility of the TCS of Cup riavidus bacteria and the excavation of their potential applications in biotechnology.
