摘 要:颗粒结合型淀粉合酶(GBSSI)是植物直链淀粉合成的关键酶,主要由W_x基因编码。本研究选用51份木薯种质资源,设计引物对W_x基因进行PCR扩增后测序。通过对获得的W_x基因序列进行了遗传多态性研究,表明其具有很大的保守性。分析W_x基因的SNP分布特点显示其主要分布在第二内含子和第四内含子之间。通过生物软件分析预测其SNP产生对颗粒结合型淀粉合酶(GBSSI)的物理化学、二级结构和三维结构的影响。本研究了解了不同种质间W_x基因的差异,可为木薯的育种提供参考依据。Granule bound starch synthase I (GBSSI) is the key enzyme of starch synthesis processes in cassava, which mainly encoded by the Wx gene. In this study, we selected 51 cassava germplasm resources and designed the primers for PCR sequencing of Wx gene. The genetic polymorphisms of the Wx gene sequences were analyzed, which indicated that it was highly conserved. Analysis of the SNP distribution of Wx gene showed that it was mainly distributed between the second intron and the fourth intron. Analysis by biological soi^are, the effects of SNP pro- duction on the physical chemistry, secondary structure and three-dimensional structure of granule-bound starch syn- thase (GBSSI) were predicted. By this study, we know the differences of IVx genes between different germplasms, which can provide reference for our cassava breeding.
