摘  要:研究运动训练对心脏衰老的保护方式.运用皮下注射D-galactose(D-gal)诱导的方式构建衰老模型,同时对衰老的小鼠进行跑步运动训练.通过实时荧光定量聚合酶链反应(quantitative reverse transcription polymerase chain reaction,q RT-PCR)技术对对照组、衰老组以及衰老跑步组小鼠的心功能指标、线粒体生成指标、能量代谢和DNA损伤指标进行检测.研究结果表明:与衰老组相比,运动组小鼠的心功能、线粒体生成能力、能量代谢水平和DNA修复能力显著提高;运动训练对心脏衰老的保护主要是通过促进线粒体的能量代谢实现的.To investigate cardio protective effects of exercise training in advanced cardiac aging. Mice were subjected to intraperitoneal injection of D-galactose (D-gal) to induce ageing, and trained using a 4-month running protocol. Heart index, mitochondrial bio- genesis, energy metabolism and DNA damage were determined by real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR). This work shows that exercise training protected heart index, increased mitochondrial DNA replication and transcription, activated the PGC-ltz signal, and decreased the DNA damage in ageing heart. In conclusion, exercise training protects against ageing-induced cardiac dysfunction through enhancing energy metabolism, improving mitochondrial biogenesis and reducing DNA damage.
