摘 要:目的观察血府逐瘀汤对动脉粥样硬化金黄地鼠主动脉的病理变化、血脂和炎症因子的影响。方法 32只雄性金黄地鼠随机分四组:正常组、模型组、血府逐瘀汤组[8.125 g/(kg·d)]、立普妥组[2 mg/(kg·d)],正常组喂食基础饲料,余组喂食高脂饲料8周,同时给予药物干预4周后处死,HE染色观察主动脉病理形态变化,ELISA检测血脂四项、血清炎症因子表达水平。结果模型组主动脉管腔狭窄、内膜增厚、泡沫细胞堆积、平滑肌细胞排列紊乱,总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白、白细胞介素-6显著升高(P<0.01);血府逐瘀汤组、立普妥组脂质斑块面积减小,三层结构较清晰,甘油三酯、白细胞介素-6、肿瘤坏死因子-α、髓过氧化物酶降低(P<0.05)。结论血府逐瘀汤可缩小斑块面积,改善血管壁结构,其作用机制可能是通过降低血脂水平,减少炎症因子的表达以减缓动脉粥样硬化病程。Objective To observe the effect of Xuefu Zhuyu decoction on the pathological changes, blood lipids and inflammatory factors in atherosclerosis golden hamster aorta. Methods 32 male golden hamsters were randomly divided into 4 groups including normal group, model group, Xuefu Zhuyu decoction group [8.125 g/(kg·d)], Lipitor group [2 mg/ (kg·d)] . Normal group was fed with basal diet. Other groups were fed with high-fat diet for 8 weeks, while were sacrificed after drug intervention for 4 weeks. HE staining was used to observe pathological changes of aorta. And ELISA was used to detect the expressions of four items of blood lipids and serum inflammatory eytokines. Results In the model group, the aortic lumen was narrow, endometrial was thickened, foam cell accumulated and smooth muscle cells arranged disorderly. And total cholesterol, triglyceride, high density lipoprotein, low density lipoprotein and interleukin-6 were significantly increased(P〈0.01). In the Xuefuzhuyu decoction group and the Lipitor group, the lipid plaque area was reduced and the three-layer structure was clear. The levels of triglyceride, interleukin-6, tumor necrosis factor α and myeloperoxidase were decreased(P〈0.05). Conclusion Xuefu Zhuyu decoction can reduce the area of plaque and improve the structure of blood vessel wall. Its mechanism may be to slow down the course of atherosclerosis by reducing blood lipid level and reducing the expression of inflammatory factors.
