摘 要:建立高效液相色谱法同时测定大黄中大黄素、大黄酚、大黄酸、芦荟大黄素、大黄素甲醚、番泻苷A、番泻苷B、没食子酸、儿茶素、(-)-表儿茶素-3-没食子酸酯、异莲花掌苷、4-4'-羟基苯基-2-丁酮、莲花掌苷、4'-羟基苯基-2-丁酮-4'-O-β-D-(2″-O-桂皮酰基-6″-O-没食子酰基)-葡萄糖苷14个成分含量的方法。采用Agilent Zorbax SB-C18色谱柱(4.6 mm×150 mm,5μm),采用0.05%的磷酸水溶液(A)-乙腈(B)为流动相,梯度洗脱,流速1.0 m L·min-1,柱温40℃,检测波长268 nm。结果表明在线性范围内14个成分线性良好(r>0.999 9);日内精密度和日间精密度均小于3.1%;平均回收率在91.80%~104.1%。同时,对收集到的10个掌叶大黄和10个唐古特大黄合格样品进行定量测定,发现掌叶大黄中含量比较高的成分是芦荟大黄素,唐古特大黄中含量比较高的是4-4'-羟基苯基-2-丁酮,各样品中所有化合物的含量差异都比较大。所建立的含量测定方法能同时测定大黄中14个成分的含量,为大黄药材多成分含量测定和质量控制提供一种简便的方法。To establish an HPLC (high performance liquid chromatography) method for the simultaneous content determination of gallic acid, (+)-catechin, (-)-epicatechin-3-O-gallate, isolindleyin, 4-(4'-hydroxyphenyl)-2-hutanone, emodin, chxTsophanol, physcion, aloe-emodin, rhein, lindleyin, 4-( 4'-hydroxyphenyl ) -2-butanone-4'-O--D-( 2"-O-galloyl-6"-O-cinnamoyl ) -glucopyrano- side, seunoside A and sennoside B in Rhei Radix et Rhizoma. The analysis was performed on Agilent Zorbax SB-CI8 (4. 6 mm 150 mm, 5 μm) with 0. 05% phosphoric acid solution (A) - acetonitrile (B) as mobile phase for gradient elution. The flow rate was 1 mL min- 1, with column temperature of 40℃ and the wavelength was set at 268 nm. All calibration curves showed good linearity (r 〉 0. 999 9 ) within the concentration range. Both the intra- and inter-day precision for 14 analytes was less than 3. 1% , with the mean re- covery at the range of 91.80%-104. 1%. Meanwhile, quantitative determination was carried out for 10 qualified samples from Rheum palmatum and 10 qualified samples from R. tanguticum, respectively. It was found that the content of 4-(4'-hydroxyphenyl) -2-buta- none and aloe-emodin were higher in the R. tanguticum and R. palmatum, respectively, and the content of all the compounds was dif- ferent in each sample. The established HPLC method for simuhaneous content determination of 14 compounds from Rhei Radix et Rhi- zoma could be used for quantitative assessment and quality control of Rhei Radix et Rhizoma.
