摘  要:目的 分析2015年长春市一起发热伴出疹疫情中患者粪便肠道病毒Echovirus 18型(E-18)毒株VP1基因序列特征及其与国内外其他流行株的进化关系.方法 采用逆转录-聚合酶链式反应(RT-PCR)扩增VP1基因片段并测序,利用DNAman软件和Mega 6.0软件进行序列分析.结果 经RT-PCR方法扩增获得4株774 bp的VP1基因片段并测序,经同源性分析比较发现,各序列间核苷酸同源性为100%.与国内外E-18病毒VP1序列同源性比较发现,长春病毒株与2015年同时发现的河北省分离株同源性最高,核苷酸同源性为97%,氨基酸同源性为99%.与Metcalf原型株比较,核苷酸同源性为80%,氨基酸同源性为93%,氨基酸对比发现有18个位点发生突变.结论 吉林省首次发现E-18型肠道病毒,为中国E-18病毒分子流行病学研究提供重要信息.Objective To analyze the genetic characters of VP1 gene of Echovirus 18 (E-18) isolated from fecal specimens of children with rush and fever in Changchun City,Jilin Province,China 2015.Methods VP1 gene of 4 samples were amplified by RT-PCR and were sequenced.The gene sequences were analyzed using DNAmanN software and Mega 6.0 software.Results In this study,VP1 genes of 4 strains were amplified from 4 samples,the length of nucleotides was 774 bp.Homology analysis indicated that the nucleotide homology of the four VP 1 gene sequences were 100%.Compared with the E-18 reference strains,Changchun strains and Hebei strains had the highest level of homology.The nucleotide sequence homology was 97%,and the amino acid homology was 99.0%.The nucleotide sequences and the amino acid sequences of Changchun strains were 80% and 93% identical to the prototype strain (Metcalf),respectively.Amino acid substitutions were found in 18 positions.Conclusions This is the first report of E-18 in Jilin Province.The study provides valuable information on the molecular epidemiology of E-18 in China.
