摘  要:以大岛樱成年优良单株‘小乔’樱的半木质化嫩枝为外植体,通过基本培养基设置、不同激素成份及浓度水平配比优化,建立其高效、稳定的离体植株再生技术。结果表明:最佳腋芽诱导培养基为:改良MS+6-BA1.5 mg/L+NAA 0.1 mg/L+蔗糖30 g/L+琼脂6 g/L,腋芽诱导快,诱导率达100%;较好的增殖培养基为:改良MS+6-BA1.0 mg/L+NAA 0.1 mg/L+蔗糖30 g/L+琼脂6 g/L,15 d增殖系数可达4.56;最适宜的生根培养基为:改良MS+NAA 1.0 mg/L+蔗糖15 g/L+琼脂6 g/L,15 d生根率达100%。炼苗后,用混合基质(泥炭土∶黄泥∶蛭石=2∶1∶1)移栽,成活率在90%以上,40 d后苗高达7~8 cm。该技术各项指标已达苗木快速繁育的要求,可直接应用于规模化生产,经济效益巨大。Study on tissue culture of C. speciosa 'Xiaoqiao' was carried out by using the late and half lignificational stems as explants. A high effective and stable plant regeneration technology of C. speciosa 'Xiaoqiao' was established through the optimization of basic medium setting, different hormone levels and composition ratio. The results showed that the optimal inducing medium was modified MS+ 6-BA 1.5 mg/L +NAA 0.1 mg/L+ sucrose 30 g/L+ Gelatin 6g/L; The optimal multiplication medium was modified MS+6- BA 1.0 mg/L +NAA 0. I mg/L+ sucrose 30 g/L+ Gelatin 6 g/L, the multiplication rate is 4.56 after 15 d; The optimal rooting medium was modified MS +NA.A 1.0 mg/L + sucrose 15 g/L+Gelatin 6 g/L, rooting rate can reach 100% within 15 d. The transplanting matrix were peat, yellow mud and vermiculite (2:1:1), and the corresponding transplanting survival rate was over 90%, the height of seedlings was up to 7-8 em 40 days after transplanting. All the indicators of this technology have reached the requirements of rapid propagation, it can be directly applied to large-scale production and will have enormous economic benefits
