摘 要:目的 将大鼠肝细胞进行传代培养 ,并观察其形态学特点 .方法 采用胶原酶二步法灌注 1 mo龄 SD雄性大鼠肝脏 ,制备肝细胞悬液并进行原代及传代培养 ,对其形态学进行了观察 .结果 肝细胞已传 7代 ,每次传代培养 6 d左右 ,光镜下可见大量新生肝细胞生长 ,呈多边形 ,相嵌紧密排列 .电镜下可见核变形 ,有的为双核细胞 .此外 ,胞质内的糖原颗粒也逐渐减少 .结论 成功进行了肝细胞的传代培养 。AIM To subculture rat hepatocytes and to observe their morphological characteristics. METHODS Hepatocytes were isolated from 1 month old rats by two step collagen perfusion. After cultured for 20 d, the primary culturesof hepatocytes were separated with trypsin and then subcultured for 7 generations whose morphological characteristics were studied. RESULTS Six days after the subculture, hepatocytes were observed under light microscope, and a great number of neoformative polygon hepatocytes were found. They were closely arranged. The nucleus of subcultured hepatocytes became deformed under TEM. Some of them were binuclear. Besides, glycogen in the cytoplasm decreased. CONCLUSION Subculture rat hepatocytes provide a convenient method for the study of hepatocytes.
