摘 要:目的:观察消渴通痹颗粒对高糖诱导的神经缺血状态下大鼠血糖及新生血管的影响。方法:将90只SD大鼠随机选出10只为空白组,其余大鼠用STZ制成糖尿病模型,3 d后凡空腹血糖≥16.7 mmol·L-1选入试验,后将其造成糖尿病周围神经病变模型。将50只造模成功的大鼠随机分为模型组、消渴通痹颗粒大剂量组、消渴通痹颗粒中剂量组、消渴通痹颗粒小剂量组、西洛他唑对照组,每组各10只。空白组及模型组均给予生理盐水0.01 mL·g-1灌胃,每日1次;消渴通痹颗粒大剂量组给予消渴通痹颗粒1.875 g·kg^(-1)灌胃,每日1次;消渴通痹颗粒中剂量组给予消渴通痹颗粒1.35 g·kg^(-1)灌胃,每日1次;消渴通痹颗粒小剂量组给予消渴通痹颗粒0.625 g·kg^(-1)灌胃,每日1次;西洛他唑对照组给予45 mg·kg^(-1)灌胃,每日1次。于末次给药后禁食12 h断尾取血,测定血糖值。同时运用HE染色法观察大鼠缺血肢体坐骨神经。结果:治疗前,与空白组比较,其他各组大鼠空腹血糖值均明显升高,差异具有统计学意义(P<0.05)。灌胃治疗后,与模型组比较,消渴通痹颗粒中剂量组空腹血糖值有所下降,差异具有统计学意义(P<0.05);与模型组比较,消渴通痹颗粒大剂量组、消渴通痹颗粒小剂量组、西洛他唑对照组空腹血糖值下降无明显变化,差异无统计学意义(P>0.05)。消渴通痹颗粒中剂量组与消渴通痹颗粒大剂量组、消渴通痹颗粒小剂量组比较,空腹血糖水平差异具有统计学意义(P<0.05)。各组大鼠缺血肢体坐骨神经组织形态学变化:消渴通痹颗粒中剂量组大鼠坐骨神经呈现个别神经纤维肿胀,鞘细胞数量少,可见坐骨神经组织血管数目有所增加。结论:消渴通痹颗粒能有效降低DPN大鼠空腹血糖,且能改善DPN大鼠缺血肢体坐骨神经组织血管数量,以消渴通痹颗粒中剂量组疗效最优。Objective:To observe the influence of Xiaoke Tongbi Granules on the blood sugar and neovascularization of rats under nerve ischemia state induced by high glucose. Methods :Ten rats among the 90 SD rats were selected randomly as blank group and the rest of the rats were made into diabetes models with STZ and made into diabetic peripheral neuropathy models. The 50 rats of successful modeling were randomized into model group, Xiaoke Tongbi Granules of high dosage group, Xiaoke Tongbi Granules of medium dosage group and Xiaoke Tongbi Granules of low dosage group and Cilostazol controt group, 10 rats in each group. Blank group and model group were given normal saline of 0.01 mL ·kg^-1for gavage, once a day;Xiaoke Tongbi Granules of high, medi- um and low dosage group were respectively given Xiaoke Tongbi Granules of 1. 875 g ·kg^-1 ,1.35 g·kg^-1 and 0. 625 g ·kg^-1 once a day;Cilostazol control group were given Cilostazol of 45 mg ·kg^-1 for garage, once a day. After 12 h of fasting after last administration, the value of blood sugar was determined. And the sciatic nerve of the ischemia limbs of the rats was observed by HE staining. Results:Before treatment,compared with blank group, the value of fasting blood sugar in the rest of the groups in- creased obviously ; the difference had statistical significance ( P 〈 0.05 ). After gavage, compared with model group, the value of fasting blood sugar in the Xiaoke Tongbi Granules of medium dosage group declined;the difference had statistical significance ( P 〈 0.05 ), while the values of fasting blood sugar in the Xiaoke Tongbi Granules of high and low dosage groups and Cilostazole group had no statistical difference (P 〉 0.05), The difference between Xiaoke Tongbi Granules of medium dosage group and Xi- aoke Tongbi Granules of high dosage group and low dosage group was significant (P 〈 0.05).
