补肾祛寒治尪汤对胶原诱导性关节炎大鼠血清IL-17、OPG及RANKL水平的影响  

Effect of Bushen Quhan Zhiwang Decoction on Serum IL-17,OPG and RANKL Levels in Rats with Collagen Induced Arthritis

摘　　要：目的:观察补肾祛寒治尪汤对胶原诱导性关节炎大鼠血清白细胞介素-17(interleukin-17,IL-17)、骨保护蛋白(osteoprotegerin,OPG)以及核因子kB受体活化因子配基(receptor activator of nuclear factor kB ligand,RANKL)水平的影响。方法:选取健康清洁级SD大鼠55只,随机选出15只作为空白对照组,其余40只全部制备胶原诱导型关节炎模型,而后随机分为模型对照组和补肾祛寒治尪汤组,每组各20只。补肾祛寒治尪汤组连续灌胃给药28 d;空白对照组正常条件饲养,自由饮水;模型对照组灌服蒸馏水。比较3组大鼠治疗后的关节炎症指数和足趾容积变化,采用酶联免疫吸附剂测定法对大鼠血清中的IL-17、OPG及RANKL水平进行测定并比较。结果:与空白对照组比较,给药1 d、5 d模型对照组和补肾祛寒治尪汤组大鼠的关节炎症指数升高,免疫后14 d足趾容积更大,且给药28 d后两组大鼠的IL-17、RANKL及OPG水平升高,差异均有统计学意义(P<0.05);与模型对照组比较,给药28 d后补肾祛寒治尪汤组关节炎症指数较低,足趾容积更小,差异具有统计学意义(P<0.05);与模型对照组比较,给药28 d后补肾祛寒治尪汤组大鼠IL-17、RANKL水平明显更低,OPG水平明显更高,差异均有统计学意义(P<0.05)。结论:补肾祛寒治尪汤可以降低胶原诱导型关节炎大鼠关节炎指数和足趾容积,其机理可能为该药降低了大鼠血清IL-17和RANKL的水平,增加OPG的血清水平。Objective： To study the effect of Bushen Quhan Zhiwang decoction on serum interleukin-17（ IL-17）,osteoprotegerin（ OPG） and receptor activator of nuclear factor kB ligand（ RANKL） levels in rat with collagen induced arthritis. Methods： 55 healthy and clean grade SD rats were selected,15 rats were randomly chosen to be the blank control group,the remaining 40 rats were all made of collagen induced arthritis model,and then they were randomly divided into model control group and Bushen Quhan Zhiwang decoction group,20 rats in each group. Bushen Quhan Zhiwang decoction group were continuous intragastrically administrated for 28 d,the blank control group was reared under normal conditions,with free drinking water,the model control group was treated with distilled water. The changes of joint inflammatory index and toe volume were compared between the 3 groups after treatment,the levels of serum IL-17,OPG and RANKL of rats were determined by enzyme linked immunosorbent assay（ ELISA）,and the result were compared. Results： Compared to the blank control group,joint inflammatory index of rats in model control groupand Bushen Quhan Zhiwang decoction group was higher at 1 d and 5 d after administration,the toe volume was larger at 14 d after immunization,and the levels of IL-17,RANKL and OPG of rats in two groups were increased at 28 d after administration,the difference was statistically significant（ P〈0. 05）. Compared with the model control group,joint inflammation index of rats in Bushen Quhan Zhiwang decoction group was lower at 28 d after administration,and the toe volume was smaller,the difference was statistically significant（ P〈0. 05）. Compared with the model control group,the levels of IL-17,RANKL of rats in Bushen Quhan Zhiwang decoction group were significantly lower,the level of OPG was significantly higher,the difference was statistically significant（ P〈0. 05）.
