摘  要:目的:观察附子-白术"药对"对成骨细胞分化和增殖的影响,并从调控BMP-2/Smads/Runx2通路方面探讨其作用机制。方法:取新生SD大鼠颅骨,多次酶消化法获得成骨细胞,分别用低、中、高剂量(1、10、100mg/m L)的附子-白术"药对"进行药物干预,MTT法检测成骨细胞的增殖,ELISA法检测成骨细胞碱性磷酸酶(ALP)活性,Western blot检测成骨细胞BMP-2、Smad1、Smad5、Runx2及Ⅰ型胶原蛋白的表达。结果:药对低、中剂量组在570nm处的吸光度值及ALP的活力均明显高于对照组(P<0.05,P<0.01),高剂量组在570nm处的吸光度值明显低于对照组(P<0.01),且高剂量组ALP的活力明显高于低、中剂量组(P<0.05,P<0.01);药对低、中剂量组成骨细胞BMP-2、Smad1、Smad5、Runx2及Ⅰ型胶原蛋白的表达均胶对照组明显升高(P<0.05,P<0.01)。结论:附子-白术"药对"可促进成骨细胞的增殖和分化,其机制可能与其激活BMP-2/Smads/Runx2信号通路,促进Ⅰ型胶原表达相关。Objective: To observe the effects of Fuzi-Baizhu couplet medicines on osteoblast differentiation and proliferation, and to explore its mechanism from the regulation of BMP-2/Smads/Runx2 pathway. Methods: Skulls of newborn SD rats were taken to be digested by multiple enzymes to obtain osteoblasts, which were intervened respectively with low, middle and high (1, 10, 100mg/mL) concentration of Fuzi-Baizhu couplet medicines. The proliferation of osteoblasts was detected by MTT, and ALP activity of osteoblasts was detected by ELISA. Expressions of osteoblast BMP-2, Smadl, Smad5, Runx2 and I collagen were detected by Western blot. Results: Fuzi-Baizhu couplet medicines on the low and middle concentration group in 570nm absorbance value and ALP activity were significantly higher than the control group (P〈0.05, P〈0.01); Fuzi-Balzhu couplet medicines on the high concentration group in 570nm absorbance value was significantly lower than the control group (P〈0.01) and its ALP activity was significantly lower than the low and middle concentration group (P〈0.05, P〈0.01); Fuzi-Baizhu couplet medicines on the low and middle concentration group of cell BMP-2, Smadl, Smad5, Runx2 and the expression of type I collagen gel were significantly higher than control group (P〈0.05, P〈0.01). Conclusion: Fuzi-Balzhu couplet medicines can promote osteoblast proliferation and differentiation, and its mechanism may be related to the activation of BMP-2/Smads/Runx2 signaling pathway and promoting the expression of type I collagen.
