摘 要:目的:通过研究中药利湿活血方及其拆方对高尿酸血症模型大鼠氧化应激损伤的影响,探讨中医临床论治高尿酸血症更为合理的治法。方法:将雄性SD大鼠随机分成空白组(10只)和造模组(65只),造模组大鼠采用酵母膏、腺嘌呤连续灌胃14d,制备高尿酸血症大鼠模型,去除未成模的5只大鼠,将造模组按血尿酸浓度随机分为模型组、四妙丸组、苯溴马隆组、利湿活血方组、拆方Ⅰ组(清热利湿药组)、拆方Ⅱ组(活血化瘀药组),每组10只。各给药组每天灌胃相应药物,连续14d。末次给药后,测定大鼠血清中血尿酸(UA)、丙二醛(MDA)含量,活性氧簇(ROS)水平及过氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)的活性。结果:与空白组比较,模型组大鼠血清UA、MDA、ROS水平显著升高(P<0.01);SOD、T-AOC活性显著降低(P<0.01)。与模型组比较,除拆方Ⅱ组外,各给药组大鼠血清UA含量显著降低(P<0.05,P<0.01);苯溴马隆组、利湿活血方组大鼠血清MDA含量显著降低(P<0.05);除四妙丸组外,各给药组SOD活性较模型组均显著升高(P<0.01);除拆方Ⅱ组外,各给药组ROS水平均显著降低(P<0.05,P<0.01);除拆方Ⅱ组外,各给药组大鼠血清T-AOC活性显著升高(P<0.05,P<0.01)。结论:中药利湿活血方在降低高尿酸血症大鼠血清UA的同时,还能增强高尿酸血症大鼠的抗氧化和清除氧自由基的能力,减少脂质过氧化,发挥综合作用,达到较好的治疗效果。Objective: Through study the effects of Lishi Huoxue Recipe onoxidative stress in hyperuricemia rats, investigate the treatment of Hyperuricemia in effective methods with Chinese medicine. Methods: Male SD rats were randomly divided into blank group (10 rats) and model group (65 rats). The model group rats were treated with yeast extract and adenine for 14 days. The 5 non-model rats were removed. The model group was randomly divided into model group, Simiaowan group, Benzbromarone group, Lishi Huoxue Recipe group, Decomposed Recipes I group, Decomposed Recipes 1/group, 10 rats per group. Each administration group was given the corresponding drugs daily for 14 days. After the last time, the UA, MDA, SOD, ROS, T-AOC, GSH-PX, CAT in serum were detected. Results: Compared with the blank group, model group rats UA, MDA, ROS level in serum were significantly increased (P〈0.01); the activity of SOD and T-AOC of model group was obviously lower (P〈0.01). Compared with the model group, each group except the Decomposed Recipes II group experimental animals serum UA content was obviously lower (P〈0.05, P〈0.01). The levels of MDA of Benzbromarone group and Lishihuoxue prescription group were significantly lower (P〈0.05). Except Simiaowan group, the SOD activity of the other groups was obviously higher than that of model group, with significant difference (P〈0.01); the ROS level of each drug group was decreased (P〈0.01). The T-AOC activity of each group except the Decomposed Recipes Ⅱ group were obviously incresaed (P〈0.05, P〈0.01). Conclusion: The Chinese herbal compounds of Lishi Huoxue Recipe reducing the hyperuricemia rat serum UA, at the same time, it can improve antioxidant and free radical scavenging capacities in hyperuricemia rats. The Chinese herbal compounds of Lishi Huoxue Recipe function could enhance curative effect, achieve the best therapeutic effect.
