摘  要:目的:研究急性脑出血应激性肝损害大鼠枯弗细胞(KCs)凋亡、核转录因子-κB(NF-κB)及肿瘤坏死因子(TNF-α)的变化,并探讨涤痰通瘀方对KCs的保护作用。方法:提取正常大鼠KCs,建立急性脑出血模型,制备正常对照组、脑出血模型组和涤痰通瘀组血清,分组给药,体外培养6、12、24h后,Western blot检测NF-κB蛋白表达,ELISA检测TNF-α含量,流式细胞技术检测KCs凋亡。结果:脑出血模型组可见较多凋亡细胞,涤痰通瘀组KCs凋亡率较脑出血模型组显著减少(P<0.05)。涤痰通瘀组在各时间点NF-κB蛋白表达水平和TNF-α含量较脑出血模型组显著降低(P<0.01,P<0.05)。结论:涤痰通瘀方能够有效抑制KCs凋亡,阻滞NF-κB的活化、减少TNF-α释放,对急性脑出血应激性肝损害具有保护作用。Objective: To study the changes of apoptosis of Kupffer cells (KCs), nuclear factor-κB(NF-κB) and tumor necrosis factor- a (TNF-α) in hepatic stress injury rats with acute cerebral hemorrhage, and explore the protective effect of Ditan Tongyu Decoction on KCs. Methods: KCs of normal rats were extracted, and the model of acute cerebral hemorrhage was established. The serum of each group (blank control group, cerebral hemorrhage group and Ditan Tongyu group) was prepared well. KCs were cultured for 6, 12 and 24h. The expression of NF-κB was detected by Western blot, the expression of TNF-α were determined by ELISA and the apoptosis of KCs was analyzed by flow cytometry. Results: There were many apoptotic cells showed in cerebral hemorrhage group, and apoptosis rate of KCs in Ditan Tongyu Decoction group was remarkably lower than that of model group (P〈0.05). Compared with cerebral hemorrhage group, the expression level of NF-κB and TNF-α in KCs in Ditan Tongyu Decoction group decreased significantly (P〈0.01, P〈0.05). Conclusion: Ditan Tongyu Decoction can inhibit apoptosis of KCs, block activation of NF-κB and reduce the release of TNF-α availably, so as to exert its protective effect on hepatic stress injury caused by acute cerebral hemorrhage.
