摘 要:目的:测定猪牙皂中刺囊酸和齐墩果酸的含量,优化猪牙皂中刺囊酸和齐墩果酸提取工艺。方法:通过单因素试验研究盐酸浓度、盐酸用量、反应温度、反应时间4个工艺参数对猪牙皂总皂苷水解的影响,以刺囊酸和齐墩果酸皂苷元的含量为指标,筛选酸水解工艺条件;采用高效液相色谱法,用十八烷基硅烷键合硅胶为填充剂,乙腈—水为流动相梯度洗脱,检测波长为215 nm,测定猪牙皂中刺囊酸和齐墩果酸的含量。结果:猪牙皂总皂苷最佳酸水解工艺条件:20%盐酸,盐酸用量80m L,水解时间2 h,水解温度90℃。在该条件下,刺囊酸和齐墩果酸的平均含量分别为62.95 mg·g-1、49.09 mg·g-1;通过方法学的系统考察,建立了猪牙皂中刺囊酸和齐墩果酸的含量测定方法。结论:本方法专属性强,重现性好,是控制猪牙皂内在质量的理想方法。In order to optimize the extraction method of echinocystic acid and oleanolic acid in Gleditsia sinensis Lam.,we studied the influence of muriatic acid concentration,muriatic acid volume,reaction temperature and reaction time on the hydrolysis of saponin through single factor test,and selected the acid hydrolysis conditions with the content of sapogenin of echinocystic acid and oleanolic acid as indexes. The content of echinocystic acid and oleanolic acid were measured by HPLC with C18 column,acetonitrile-water as the mobile phase and the detection wavelength at 215 nm. The results showed that the optimum acid hydrolysis conditions were as follows: 80 mL 20 % muriatic acid,hydrolysis at 90 ℃ for 2 h. Under the above conditions,the average content of echinocystic acid and oleanolic acid reached up to 62. 95 mg·g^(-1) and 49. 09 mg·g^(-1) respectively. This method has the advantages of high specificity and good repeatability,and is the ideal method to control the quality of Gleditsia sinensis Lam.
