摘　　要：目的:利用伏安行为的电化学方法研究Balb/c3T3细胞转化过程中嘌呤核苷酸代谢变化规律,确定细胞转化的电化学检测指标。方法:利用MNNG与TPA诱导Balb/c3T3细胞体外进行转化,同时利用ITEs进一步加速转化效率,在不同时间点对转化的细胞进行细胞形态学观察、软琼脂克隆实验及伏安行为电化学检测,电化学法跟踪BALB/c3T3细胞转化过程中嘌呤含量的变化。结果:实验第10天开始,转化细胞的嘌呤的信号峰值明显高于对照组(DMSO组),细胞内嘌呤含量增多,同时细胞增殖加速。ITEs可缩短转化时间。结论:嘌呤可作为电化学法检测的生物标记物,伏安行为的的电化学法可以快速检测体外细胞发生恶性转化的过程,与传统方法相比,缩短了检测周期,有望成为一种简单、快速、灵敏、低成本的细胞转化检测新手段。Objective: To study the metabolism variation regularity of purine nucleotide and ensure the electrochemical biomarkers during the process of Balb/c3 T3 cells transformation by using electrochemical detection methods. Methods: MNNG and TPA were used to induce the transformation of Balb/c3 T3 cells in vitro. The transformation efficiency was further accelerated by ITEs. At different time,the transformed cells were observed by cell morphological and soft agar cloning methods. The changes of purine level in BALB/c3 T3 cells were detected by electrochemical method. Results: From the 10 th day of the experiment,the peak value of purine signal began significantly higher than that of the control group( DMSO group). The intracellular purine content began to increase,cell proliferation rate was accelerated,and conversion time was shortened by ITEs. Conclusions: Purine can be used as biomarker of electrochemical detection; Voltammetric behavior of the electrochemical method can quickly detect the malignant transformational process of cells in vitro; Compared with the traditional method,voltammetric behavior of electrochemical method shortened the detection period,and was expected to become a simple,sensitive,low-cost new method for cell transformation test.
