摘 要:目的:对比三种高尿酸血症小鼠模型的制备方法,以期建立稳定的慢性高尿酸血症小鼠模型。方法:分别采用酵母+腺嘌呤、腺嘌呤+乙胺丁醇和腺嘌呤+氧嗪酸钾三种药物组合进行造模,灌胃给药;检测小鼠的血尿酸、肌酐等生化指标;石蜡包埋制作病理切片,应用苏木精-伊红染色观察肾脏病理变化。结果:试验第14天开始,各模型组小鼠的体重均下降,尤以酵母+腺嘌呤组下降最明显,且血清中尿酸、血肌酐和血尿素等生化指标呈现明显的升高趋势。结论:通过酵母+腺嘌呤建立的高尿酸血症小鼠模型的生化指标、病理变化更显著,该模型更接近于人高尿酸血症的病理变化。Objective: Compare three methods for hyperuricemia mouse models in order to establish a stable chronic hyperuricemia mouse model. Methods: The model was made by two combinations of yeast,adenine and ethambutol respectively. Drugs were administered orally to the mice. The serum uric acid,creatinine and other biochemical indexes were detected. Pathological changes were observed by hematoxylin eosin staining in paraffin embedded sections. Results: At the beginning of the fourteenth day,the body weight of the mice in the model group decreased,especially in the yeast + adenine group,and the serum uric acid,serum creatinine and blood urea showed an obvious upward trend.Conclusion: The changes of biochemical indexes and pathology in hyperuricemia mouse model established by yeast and adenine were more obvious,so that the model established by yeast and adenine was more close to the pathological changes of hyperuricemia.
