摘  要:目的:研究连翘酯苷A和内毒素作用鸡后,鸡脾器官指数、鸡血IL-17和IL-6 mRNA表达量与蛋白含量与炎症反应关系。方法:选取80只1d龄健康AA肉鸡随机分为4组,分别为control组,LPS组,30组(30 mg·kg^(-1)连翘酯苷低剂量组)和60组(60mg·kg^(-1)连翘酯苷高剂量组),各组鸡收集鸡脾脏、血和血清,计算鸡脾脏器官指数、通过采用qRT-PCR和ELISA方法检测血液和血清中IL-17和IL-6水平。结果:与正常组相比,LPS组脾器官指数上升,血液中IL-17和IL-6 mRNA表达量升高;与LPS组比较,连翘酯苷A剂量组脾器官指数下降,血液中IL-17和IL-6 mRNA表达量下降;与正常组相比,LPS组血清中IL-17和IL-6蛋白含量升高;与LPS组比较,连翘酯苷A组血清中IL-17和IL-6蛋白含量下降。结论:试验表明连翘酯苷A可以通过基因和蛋白水平抑制LPS诱导的鸡血中IL-17和IL-6 mRNA表达量和蛋白含量的升高,起到免疫调节作用,达到抗炎效应。Objective: To investigate the relationship among spleen organ index,inflammatory reaction,IL-17 and IL-6 protein and mRNA levels in blood of chicken after administrated by Forsythiaside A and Lipopolysaccharide( LPS). Methods: 80 AA broilers were selected and randomly divided into four groups,control group,LPS group,low Forsythiaside A dosage group and high Forsythiaside A dosage group. Spleen、blood and serum were collected for IL-17 and IL-6 level detection by qRT-PCR and ELISA. Results: Spleen organ index,IL-17 and IL-6 mRNA blood levels in LPS group were increased compared with control group. Spleen organ index,IL-17 and IL-6 mRNA expression in Forsythiaside A groups were lower than LPS group. IL-17 and IL-6 mRNA expression in chicken blood were enhanced which was induced by LPS. Decreased protein expression of IL-17 and IL-6 in serum were observed in Forsythiaside A groups,as compared with LPS group. Conclusions: Forsythiaside A achieved its immunomodulatory and anti-inflammatory effect by inhibiting the increased IL-17 and IL-6 expression induced by LPS in chicken.
