废次烟叶多糖绿色提取工艺的研究

摘　　要：废次烟叶富含多糖具有十分重要的开发价值。文章研究了热水浸提烟叶多糖的提取工艺,考察浸提料液比、时间、温度和浸提剂乙醇体积分数对烟叶多糖提取率的影响,在其基础上进行正交试验。结果表明,影响热水浸提烟叶多糖提取率的主次因素依次为浸提剂乙醇体积分数、温度、料液比、浸提时间。热水浸提烟叶多糖的最优组合条件为:浸提剂为蒸馏水,温度为80℃,料液比为1∶60,时间为30min。上述条件下连续提取2次,多糖提取率可达4.08%。在该工艺条件下,烟叶多糖的提取率与微波提取法相当。该工艺不涉及微波、超声波等辅助设备和有机溶剂的使用,具有操作方便、成本低、环保等优势。Waste tobacco leaves,which are rich in polysaccharides,have very fundamental development value.In this paper,the extraction technology of polysaccharides from tobacco leaves by hot water extraction was studied,the effect of the ratio of raw tobacco to extraction medium,time,temperature and ethanol concentration of extraction medium on polysaccharides yield was investigated,and then the orthogonal test was conducted.The results showed that the importance for polysaccharides yield decreased in the following order:ethanol concentration of extraction medium,extraction temperature,ratio of raw tobacco to extraction medium and extraction time.The optimum conditions for polysaccharides extraction were established as follows:the extraction medium was distilled water,the extraction temperature was 80°C,the ratio of raw tobacco to extraction medium was 1∶60 and the extraction time was 30 min.The total polysaccharides yield of twice extraction was up to 4.08% under the presented conditions,which was similar to that under microwave-assisted extraction.This process does not involve in the use of microwave or ultrasonic equipment and organic solvent.So it has the advantages of convenient operation,low cost and environment-friendliness.
