摘 要:目的:研究甘草酸对UVB(中波紫外线)诱导人皮肤角质形成细胞(Ha Ca T细胞)凋亡的影响。方法:以不同浓度的甘草酸作用于Ha Ca T细胞,噻唑蓝比色(MTT)法筛选药物的有效安全浓度;用总剂量为30 m J/cm-2的UVB照射Ha Ca T细胞,制备光老化模型,MTT法检测细胞增殖率;流式细胞仪检测各组细胞总凋亡率;实时定量PCR(RT-PCR)及蛋白免疫印迹(Western Blot)法分别检测Caspase-3、Caspase-9 m RNA和蛋白表达水平。结果:1×10-5、1×10-6、1×10-7mol/L甘草酸为最佳有效安全浓度。与空白组比较,UVB组细胞增殖率显著降低(P<0.01),细胞总凋亡率显著显著升高(P<0.01),Caspase-3、Caspase-9 m RNA及蛋白表达水平显著升高(P<0.01)。与UVB组比较,1×10-5、1×10-6、1×10-7mol/L甘草酸+UVB组细胞增殖率显著升高(P<0.01),细胞总凋亡率显著显著降低(P<0.05,P<0.01),Caspase-3、Caspase-9 m RNA及蛋白表达水平显著降低(P<0.05,P<0.01)。结论:甘草酸能抑制UVB诱导的Ha Ca T细胞凋亡,其机制与其降低细胞总凋亡率,抑制Caspase-3、Caspase-9 m RNA和蛋白的表达有关。Objective To study the effects of glycyrrhizic acid on ultraviolet B(UVB)induced apoptosis of human keratinocytes(HaCaT cells).Methods The HaCaT cells were treated by different concentrations of glycyrrhizic,the effective and safe concentration of glycyrrhizic acid was screened by the method of methyl thiazolyl tetrazolium(MTT).Photoaging model was set up by the irradiation of ultraviolet B(UVB)at irradiation dose of 30mJ/cm2,the activity of cells was determined by MTT assay.The apoptosis of HaCaT cells were respectively measured by flow cytometry.The expression of Caspase-3 and Caspase-9 mRNA and protein were measured by Real-time Quantitative PCR(RT-PCR)and Western Blot.Results The best safe and effective concentration of glycyrrhizin were 1×10-5,1×10-6,1×10-7mol/L.Compared with the blank group,the cell proliferation rate of the UVB group was significantly decreased(P〈0.01),the total apoptosis rate of cells was significantly increased(P〈0.01),the expression of Caspase-3 and Caspase-9 mRNA and protein were significantly increased(P〈0.01).Compared with the UVB group,the cell proliferation rate of the(1×10-5,1×10-6,1×10-7)mol/L glycyrrhizic acid+UVB group were significantly increased(P〈0.01),the total apoptosis rate of cells was significantly decreased(P〈0.05,P〈0.01),the expression of Caspase-3 and Caspase-9 mRNA and protein were significantly decreased(P〈0.05,P〈0.01).Conclusion Glycyrrhizic acid can inhibits the apoptosis of HaCaT cells induced by UVB,the mechanism can be related with decreaseing the total apoptosis rate of cells,inhibiting the expression of the Caspase-3 and Caspase-9 mRNA and protein.
