摘 要:目的通过采用RT-PCR方法观察降钙素基因相关肽(CGRP)在正常股骨头组织与缺血坏死性股骨头组织中基因的表达差异,探讨CGRP在股骨头坏死中的发病机制。方法分别取正常股骨头、缺血坏死性股骨头骨组织各12例;分别取骨组织3~4g放入冷冻管存入液氮,用于CGRP基因的RT-PCR。结果 CGRP在正常骨组织中的表达量比其在坏死组织中明显减少,RTPCR的2^(-ΔΔCt)平均值分别为(0.42±0.24)和(0.53±0.35);正常股骨头骨组织中CGRP基因RTPCR的2^(-ΔΔCt)平均值与缺血坏死性股骨头骨组织平均值相比,P<0.05。结论 CGRP基因在正常与缺血坏死性股骨头骨组织中的表达量明显不同,因此CGRP基因表达量的增多也许是股骨头缺血坏死的重要原因之一。Objective The difference of gene expression between calcitonin gene related peptide(CGRP)in normal femoral head and ischemic necrotic femoral head was observed by RT-PCR method,and the pathogenesis of CGRP in osteonecrosis of femoral head was discussed.Methods 12 cases of normal femoral head and ischemic necrosis of femoral head were selected,respectively.Bone tissue 3~4 g was injected into cryopreserved tube into liquid nitrogen for CGRP gene RT-PCR.Results The expression of CGRP in normal bone tissue was significantly lower than that in necrotic tissue,and the average value of RT-PCR 2^(-ΔΔCt) was(0.42 + 0.24)and(0.53 + 0.35),respectively.The average value of 2^(-ΔΔCt) of the CGRP gene RTPCR in the bone tissue of the normal femoral head was compared with the mean value of the ischemic necrosis of the bone tissue of the femoral head,P <0.05.Conclusion The expression of CGRP gene in normal and ischemic necrotic femoral head bone tissues is significantly different.Therefore,the increase of CGRP gene expression may be one of the important reasons for avascular necrosis of femoral head.
