摘 要:目的研究氧化应激与2型糖尿病患者发病程度的关系。方法通过电子自旋偶联共振(ESR)技术检测各组血清活性氧(reactive oxygen species,ROS)水平,分析2型糖尿病患者与正常人血清氧化应激水平差异,以及2型糖尿病患者间氧化应激水平与发病程度之间的关系;通过实时荧光定量RT-PCR技术分析与氧化应激水平相关的基因的表达水平,在基因层面分析氧化应激水平与2型糖尿病患者发病程度之间的关系。结果与正常组相比,2型糖尿病患者组ROS显著上升(P<0.01),氧化应激相关基因乙醛还原酶1基因、谷胱甘肽过氧化物酶1基因、β-丙氨酸丙酮酸转氨酶基因、含黄素单氧化酶5基因均显著下调(P<0.01)。结论 2型糖尿病患者随着发病程度加深,空腹血糖水平上升,并伴随着ROS含量的增加,最终导致氧化应激相关基因表达下调。Objective To study the relationship between oxidative stress and the incidence of type 2 diabetes mellitus. Methods The serum reactive oxygen species( ROS) level was detected by means of electron spin resonance coupling( ESR),then the differences of serum oxidative stress level between patients with type 2 diabetes and normal people and the relationship between the level of oxidative stress and the severity of type 2 diabetes were respectively analyzed. The expression level of oxidative stress-related genes was detected using real-time fluorescence quantitative( RT-PCR),and the relationship between the severity of type 2 diabetes and oxidative stress level was analyzed at the genetic level. Results The ROS level significantly increased in type 2 diabetes group than the normal group( P〈 0. 01),and oxidative stress-related genes such as aldehyde reductase 1 gene,glutathione peroxidase 1 gene,β-alanine pyruvate transaminase gene,flavin-containing monooxygenase 5 gene significantly decreased than the normal group( P〈 0. 01). Conclusion The levels of fasting blood-glucose and ROS of patients of type 2 diabetes rise with the deterioration of disease,which leads to the downregulation expression of oxidative stress-related genes.
