摘 要:以湖羊为研究对象,采用RT-PCR法对IGF-Ⅰ和MyoG在湖羊不同生长阶段背最长肌中的表达丰度及其与屠宰性状的相关性进行研究,以揭示湖羊肌肉生长性状的分子遗传学基础。结果表明:(1)随着年龄的增加,母羊IGF-Ⅰ在背最长肌中的表达趋势,由2日龄开始逐渐升高,并于6月龄达最高点;公羊IGF-Ⅰ在背最长肌中的表达趋势,先升高至1月龄时达一峰值,1~3月龄为一个降低的过程,且于3月龄降至与初生相近水平,此后随着年龄的逐渐增加,于6月龄达最高点。母羊MyoG在背最长肌中的表达趋势,先升高后降低再升高再降低最后再升高,并于6月龄达最高点;公羊MyoG在背最长肌的表达趋势,2日龄至1月龄处于上升过程,1~4月龄为下降过程,4~6月龄又逐渐上升,且于6月龄达最高点。IGF-Ⅰ和MyoG在湖羊各月龄间大多存在显著或极显著差异,公羊和母羊在相同生长阶段IGF-Ⅰ和MyoG的表达大多存在显著或极显著差异。(2)IGF-Ⅰ和MyoG表达呈极显著正相关,IGF-Ⅰ与宰前活重呈极显著正相关,与胴体重呈显著正相关,与净体重呈正相关。这一研究显示,IGF-Ⅰ和MyoG基因存在极显著正相关,受年龄的影响对湖羊早期肌肉生长发育具有重要的促进作用,且在湖羊早期肌肉生长发育过程中可能存在性别特异性。In this study, Hu sheep was researched object and methods of RT-PCR was applied to analyze the expres sion of IGF- ~ and MyoG in Hu sheep longissimus dorsi at different growth stages and their association analysis with meat quality traits, and the relationship of the expression level between IGF- I and MyoG. The objective was is to reveal the genetic background of muscle growth in Hu sheep by using candidate gene method and provide genetic information for the selection of muscle growth in Hu sheep. The results were as follows: (1) With increasing age after birth, the IGF- I expression trend of ewes was increasing from 2-day to 6 month which had the top expression; the IGF I expression trend of rams was that 2-day firstly increased to a peak at one-month, 1-month to 3-month had a decreasing process and reached the same level which was close to the newborn, and then inceased with the increasing age, 6-month reached the top. With increasing age after birth, the MyoG expression trend of ewes was up-down-up-down-up, and 6-month reached the top; the MyoG expression trend of rams was that 2 day firsdy increased to a peak at one-month, 1-month to 4-month was a process of decreasing, 4 month gradually increased to 6-month which reached top expression. Most growth phases had the significant difference or extreme significant difference in IGF- I and MyoG and the expressions of IGF- T and MyoG between most rams and ewes that at the same growth phase had the significant difference or extreme significant difference. (2) The expressions of IGF- I and MyoG had an extreme significant positive correlation, the expressions of IGF- I had an extreme significant positive correlation with live weight and a significant positive correlation with carcassweight and had no significant correlation with net meat weight; the expression of MyoG had no significant correlation with live weight, carcass weight and net meat weight. The results showed that IGF- I and MyoG had an extreme significant positive effect on Hu sheep early muscles
