摘  要:目的:建立喜树叶中喜树碱(CPT)含量测定的HPLC方法,并对喜树嫩叶和成熟叶中CPT含量的季节变化以及不同树龄喜树叶片中CPT含量进行测定。方法:应用HPLC法测定喜树嫩叶与成熟叶中CPT含量,对喜树嫩叶与成熟叶中CPT含量季节性变化规律进行研究,对不同树龄喜树叶片中的CPT含量进行比较,同时对测定方法的可靠性进行评价。结果:建立的喜树嫩叶与成熟叶中CPT含量测定方法准确可靠,且在生长期内,无论是嫩叶还是成熟叶,CPT含量均为单峰曲线,均在7月中旬含量达到最大值;且嫩叶中CPT含量均高于同期成熟叶;树龄不同,喜树叶片中CPT含量亦不相同,树龄越大,叶片中的CPT含量越低。结论:HPLC法可作为喜树叶片中CPT含量的测定方法,且实验结果准确可靠。在生长期内,喜树嫩叶与成熟叶中CPT含量季节变化趋势呈单峰曲线变化,含量最高值均出现在7月中旬,嫩叶中CPT含量较同期成熟叶高;喜树叶片中的CPT含量随着树龄的增大而降低。Objective:To determine the seasonal changes of camptothecin(CPT)content in young and mature leaves of C. acuminate with HPLC method and to compared the CPT content in leaves of C. acuminata at different ages. Methods:The content of CPT in young and mature leaves of C. acuminata was determined by HPLC method,the seasonal changes of CPT content in young and mature leaves of C. acuminata was studied,and CPT content in leaves of C. acuminata at different ages was compared. The reliability of the method was evaluated. Results:The determination of CPT content in young and mature leaves of C. acuminata by HPLC was accurate and reliable. The contents of CPT were a single peak curve in both young and mature leaves,and reached the maximum value in the middle of July. The content of CPT in young leaves was higher than that in mature leaves at the same period.The greater age of C.acuminata had the lower content of CPT in leaves. Conclusions:The HPLC method can be used as a method to determine the contents of CPT in leaves of C. acuminata,and the experimental results are accurate and reliable. During the growth period,the seasonal variation trend of CPT content in young and mature leaves of C. acuminata show a single peak curve,the highest content appeares in the middle of July. The CPT content in young leaves is higher than that in mature leaves at the corresponding stage. The CPT content in leaves of C. acuminata decreases with the increase of tree age.
