摘 要:目的:探讨一氧化氮供体S-亚硝基谷胱甘肽(GSNO)对储存期内机采血小板代谢的影响。方法:随机采集机采血小板6份,每份150~170ml,混匀后无菌分装于血小板专业保存袋中,共5袋,每袋30~35ml,4个试验组(向各组中每1袋血小板加入不同浓度GSNO溶液5ml,最终浓度依次为5、10、100和200μmol/L)和1个对照组(加入等量的无菌生理盐水),于(22±2)℃震荡保存7d,分别在第1、3、5、7天取血小板2ml进行血小板常规指标、血气分析、MTS等指标检测。结果:对照组NO浓度随时间增加而小幅增长,试验组中,高浓度组(100、200μmol/L)在第3天达到顶峰,低浓度组(5、10μmol/L)在第5天达到顶峰。血小板pH值在第7天保存期中呈下降趋势,且保存至第7天时仍符合血小板储存期末pH值要求。2组pH、MPV、PDW、pCO2、pO2、cHCO3等指标比较差异无统计学意义(P=0.07,P=0.47,P=0.59,P=0.15,P=0.64,P=0.06),随时间增长对照组和处理组pH、pCO2、cHCO3均逐渐降低趋势,而pO2L、MPV、PDW呈逐渐升高趋势。随GSNO浓度增大,Lac含量递减;随时间增长,Lac含量逐渐增加。保存期内血小板2组MTS活性随时间增长而递减,GSNO处理的血小板与对照组相比均差异无统计学意义(P=0.18,P=0.08,P=0.24,P=0.51)。结论:血小板保存过程中加入NO供体GSNO,一定程度上可以抑制血小板代谢,减少乳酸形成,改善血小板功能。Objective:To investigate the effect of nitric oxide(NO)donor S-nitrosoglutathione(GSNO)on the metabolism of aphaeresis platelets during storage.Method:Randomly collected six copies of aphaeresis platelet with each about 150-170 ml were mixed and separated into five professional storage bags with each about 30-35 ml.Five groups were consisted of four experimental groups(added with different concentrations of 5 mL GSNO respectively and the final concentration were 5,10,100,200μmol/L)and one control group(added with equal amount of normal saline).The samples were shaked and saved for 7 dat(22±2)℃ and 2 ml platelet were used to count and detection of pH,Lac and MTS at d1,d3,d5 and d7.Result:NO concentration in control group increased slightly with time,while in the experimental groups,high concentration groups(100,200μmol/L)reached the peak at d3 and low concentration groups(5,10μmol/L)peaked at d5.During 7 dstorage period,the pH value of platelets showed a downward trend and met the requirements at d7.The values of pH,MPV,PDW,pCO2,pO2 and cHCO3 had no differences between treatment and control groups(P=0.07,P=0.47,P=0.59,P=0.15,P=0.64 AND P=0.06).With time,the values of pH,pCO2 and cHCO3 in treatment and control groups decreased gradually,while the values of pO2 L,MPV and PDW showed a gradually increasing trend.Lac content decreased with the increasing of GSNO concentration and the content of Lac increased gradually with time.MTS activity of platelets decreased with time in the experimental and controls groups during the preservation period.Platelets with GSNO treatment had no significant differences compared with the control group(P=0.18,P=0.08,P=0.2414,P=0.51).Conclusion:In the preserved process of platelets,NO donor GSNO could inhibit platelet metabolism,reduce lactic acid formation and improve platelet function to some extent.
